摘 要:通过RT-PCR分段扩增PRRSV流行株Jiangxi-3的基因组,参照VR2332序列,根据各片段之间相重叠的区域,将扩增片段的序列依次拼接获得Jiangxi-3完整的全基因组,并通过DNAStar软件对其进行分析。结果表明:Jiangxi-3的Nsp2推导氨基酸发生30个氨基酸的不连续缺失,与HB-1(sh)/2002相比,Nsp2推导的第480位氨基酸由D突变为E,缺失的位置为第481位氨基酸和532~560位氨基酸;Jiangxi-3与2006年Tian Kegong等从江西"高热病"病死猪体内分离的毒株JXA1的各个结构蛋白阅读框的氨基酸同源性为98.0%~100%;Jiangxi-3与HB-1(sh)/2002、VR2332、RespPRRSMLV的Nsp2推导的氨基酸同源性分别为94.7%、77.8%、77.7%,由遗传系统发育树可知与2006年以前分离的毒株相比,Jiangxi-3与HB-1(sh)/2002的亲缘关系较近。说明Jiangxi-3和JXA1极可能由同一毒株HB-1(sh)/2002演化而来。Whole-genome sequence of isolate Jiangxi-3 were obtained by splicing of many RT-PCR fragments according to the sequence of VR2332. Whole-genome analysis revealed that Jiangxi-3 isolate are highly homologous to HB-1(sh)/2002, a Chinese strain of PRRSV (94.7% amino acids identity of Nsp2). More importantly, we observed a unique molecular hallmark in the viral isolate, namely a discontinuous deletion of 30 amino acids in nonstructural protein 2 (Nsp2). Compared with HB-1(sh)/2002, 480 aa was mutated (D→E) and the position of deletion were 481 aa and 532-560 aa. Compared with JXA1 isolate 98.0%-100% homology for every gene reading frame, what is a Chinese strain of PRRSV isolated from "high fever" disease in pig herds by Veterinary Diagnostic Department China Animal Disease Control Center in 2006. Jiangxi-3 shared 94.7%, 77.8%, 77.7% amino acid homology for Nsp2 compared with HB-1 (sh)/2002, VR2332, RespPRRS MLV. The results showed that Jiangxi-3 and JXA1 were probably evolved from HB- 1 (sh)/2002.
