摘 要:目的观察单眼剥夺大鼠神经生长因子(nerve growthfactor,NGF)治疗前后外侧膝状体背核(dorsal lateral geniculate nucleus,dLGN)神经型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)阳性神经元表达的改变。方法健康雄性SD大鼠,随机分为正常组(N)、单眼剥夺组(MD)、单眼剥夺脑室内注射NGF组(MD+NGF)和注射生理盐水组(MD+NS),每组随机分为2个亚组。剥夺自14 d龄开始,分别于28、42 d龄处死,脑室注射分别于14、28 d龄开始。采用SABC免疫组织化学染色法观察dLGNnNOS阳性神经元的表达。结果nNOS阳性神经元在dLGN散在分布,单眼剥夺和年龄增长可致其阳性神经元数密度降低。N组及MD+NGF组分别与MD组和MD+NS组比较差异均有统计学意义(P<0.05)。42 d龄处死大鼠dLGN阳性神经元数密度N组与其他3组比较差异有统计学意义(P<0.05)。4组42 d龄与28 d龄比较,dLGN阳性神经元数密度均有显著下降(P<0.05)。结论视觉发育具有可塑性,弱视治疗具有时效性。在视觉发育敏感期内,外源性提供NGF,可以促进dLGNnNOS增多,从而拮抗剥夺效应。Objective To observe the effect of NGF on the expression of nNOS in dorsal lateral geniculate nucleus (dLGN) of monocular deprived rats. Methods The healthy male SD rats were randomly divided into four groups: normal group (N), monocular deprivation group (MD) and monocular deprivation combined with intraventricular injection of β-NGF group (MD+ NGF) or NS group (MD + NS). The rats were deprived in left eyes, beginning from postnatal day 14. Each group was divided into two subgroups, in which the rats were killed at postnatal day 28 (subgroup 1) and 42 (subgroup 2) respectively. The intubation tube in right ventricule was performed at postnatal day 14 in the last two groups, in which injections(2 μl) were given for 2 weeks, and the subgroup 1 was injected beginning from postnatal day 14 and the subgroup 2 was injected beginning from postnatal day 28. By using the anti-nNOS polycloned antibodies immunocytochemical staining, the changes of numberal density of neuronal nitric oxide synthase (nNOS) positive neurons in dLGN were studied. Results The nNOS immunopositive neurons appeared brown colour were scattered in dLGN. The numberal density of immunopositive neurons decreased along with age and by MD. It increased in MD + NGF subgroup 1 comparing with MD group, and so does in N group. But the same situation were not seen in MD + NGF subgroup 2. Conclusion The visual development is fictile and the therapy to amplyopia have chronergy. In sensitive period of visual development, the ectogenic NGF was effective in preventing the effect of monocular deprivation in rat through the increased nNOS in dLGN.
