摘 要:以芦丁为标准品,采用分光光度法,测定总黄酮的含量,对波长、精密度、重现性和稳定性进行了检测。并且利用微波辅助萃取技术,对鹰嘴紫云英、红豆草、百脉根和小冠花4种豆科牧草的总黄酮进行了提取。结果表明,此方法简便易行,精密度高,重现性好,在20-40 min,吸光度较为稳定。这4种牧草均含有丰富的黄酮类物质,且茎、叶、花、全株总黄酮含量有极显著差异(P〈0.01),在整个生育时期,总黄酮含量均为叶高于茎。在分枝、孕蕾、初花、盛花和结荚表现出不同的变化规律。小冠花和百脉根总黄酮含量的变化相近,随植株生长而不断升高,在盛花期达到最高。鹰嘴紫云英的总黄酮含量呈“V”型变化,初花期最少,只有8.71 mg/g。红豆草叶的总黄酮含量呈缓慢升高,结荚期最高。The colorimetric method was used for quantification of flavonoid using rutin as standard reagents, which detected on wavelength, accuracy, reproducibility and stability. The total flavonoid in four species of legumes (Astragalus sinicus, Onobrychis viciaefolia, Lotus corniculatus, Coronilla varia ) were extracted by microwave assistant respectively. The results showed that this method was simple, it had high accuracy, repro- ducibility and excellent stability from 20 min to 40 min; these four legumes were abundant in flavonoid. The contents of total flavonoid had significant differences (P〈0. 01) in stems, leaves, flowers and the whole plant, and that in leaves were much higher than in stems of four species of legumes in the whole growing period. The changing trend show difference in shooting stage, bud stage, early flowering, flowering and pod stage. It also showed that the changes of contents in Lotus corniculatus was the same with that in Coronilla varia, Both of them had the highest total flavoniod contents in flowering stage. The changes of total flavoniod contents in Astragalus sinicus presented "V" shape with the lowest content in early flowering. The total flavoniod contents in Onobrychis viciaefolia slowly increased and reached the highest content in pod stage.
