玉米花粉总RNA提取方法的比较和分析

摘　　要：以玉米花粉为材料,借鉴其他植物RNA的提取方法,比较了利用Trizol法、酸性酚—异硫氰酸胍法和CTAB改进法提取玉米花粉RNA的效果,利用普通琼脂糖凝胶代替甲醛变性胶电泳分析3种不同方法所提取的RNA的完整性,并用紫外分光光度计分析RNA的纯度。结果表明,CTAB改进法为其中最好的方法。The maize pollens were used to extract total RNA using 3 different methods （Trizol, acid guanidinium isothiocyanate-phenol-ehloroform and modified CTAB） and their extraction effects were compared,with reference to the RNA isolation method for other plants. The agarose gel electrophoresis,instead of formaldehyde denaturalization electrophoresis, was used to analyze the integrality of RNA extracted by 3 methods,and the UV-spectrophotometer was used to analyze the purity of RNA. The result showed that the modified CTAB method was the best.
