摘  要:目的对南蛇藤中扁蒴藤素提取纯化工艺进行研究。方法南蛇藤根粉经丙酮冷浸提取后,采用聚酰胺柱层析及硅胶柱层析进行分离纯化;以HPLC法测定产品中扁蒴藤素的含量,并以其为指标对分离纯化的主要工艺参数进行优化。结果将待分离的南蛇藤丙酮浸膏和聚酰胺(60~100目)按物料比0.02(质量比)装入层析柱中,常压下用体积分数75%的甲醇洗脱至洗脱液近无色,收集流分,浓缩后得到粗品,经硅胶柱层析进一步纯化,再经重结晶后纯度提高到98.0%以上。结论该工艺简单、成本较低,适于工业化。Objective To study the isolation and purification condition of pristimerin from Celastrus orbiculatus Thunb. Methods Pristimerin was isolated and purified by polyamide column chromatography and silica gel column chromatography, and the content of pristimerin was determined by HPLC. The main isolation factors were optimized. Results The optimum isolation conditions were as following: the weight ratio of sample to polymide was 0.02, 75% methanol was used for eluent. Under the conditions above, crude product was obtained, which was further purified by silica gel column chromatography and recrystallized. Finally, the purity reached to 98%. Conclusion This method is simple and economic, which could be used in industry.
