摘 要:[目的]研究羟基红花黄色素A(Hydaosafflow yellow A)对一次性力竭游泳运动后大鼠大脑皮质血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)及其受体2(kinase insert domain containing receptor,Flk-1/KDR)表达的影响,探讨羟基红花黄色素A的脑保护作用机制。[方法]采用动物实验,SD大鼠随机分为空白对照组、生理盐水组(PSS组)和羟基红花黄色素A组(HYA组)。适应性喂养1周后,运动前30min分别腹腔注射生理盐水和羟基红花黄色素A,进行一次性力竭运动,分别在力竭运动后即刻、12和24h灌注取材,采用免疫组织化学检测大脑皮质VEGF和KDR的表达。[结果]VEGF和KDR的阳性表达位于神经元的细胞浆和细胞膜;PSS组和HYA组在力竭即刻出现VEGF和KDR的表达增强;12h下降;24h又有所回升,尤以KDR表达更为显著(P﹤0.05);在力竭运动后即刻和24h,HYA组VEGF和KDR的表达均高于PSS组;差异均有统计学意义(P值均﹤0.05)。[结论]一次性力竭运动后VEGF和KDR动态变化具有时间和空间的一致性,羟基红花黄色素A可能通过上调神经元VEGF和KDR的表达,以自分泌机制对神经元起到保护作用。[Objective] To investigate the influences of Hydaosafflow yellow A on Vascular Endothelial Growth Factor (VEGF) and kinase insert domain containing receptor (KDR) in rats cerebral cortex after exhaustive swimming, exploring the meaning of those changes and the mechanism of the protective effect. [Methods] Rats were selected and divided randomly into control group, physiological saline solution group (PSS group) and Hydaosafflow yellow A (HYA group). All the rats were fed adaptively for 1 week. Rats in PSS group and HYA group were injected (IP) physiological saline solution and Hydaosafflow yellow A respectively 30 min before swimming, and then forced to one-time exhaustive swimming. Materials were drawn from rats in the instant time 12h and 24h later with 4% paraformaldehyde by filling. Cerebral cortex was gained to perform VEGF and KDR immunohistochemical staining. [Results] The positive expression of VEGF and KDR located in cell plasm and cell membrance. The expression of VEGF and KDR in both PSS and HYA groups in the instant time was stronger (P﹤0.05), went down 12h later (P﹤0.05), and up again 24h later, especially the expression of KDR (P﹤0.05). The expression of VEGF and KDR in HYA group was higher than that in PSS group in the instant time and 24h (P﹤0.05). [Conclusion] The temporal and spatial expression pattern of VEGF correlates with the expression of KDR after exhaustive exercising. The rapid and temporary expression of VEGF / KDR after exhaustive exercising seem not to form new blood vessel, but to play a part in neuroprotection. Hydaosafflow yellow A could reduce the damage of the brain after exhaustive swimming by upregulation of VEGF and KDR in neuron.
