摘  要:[目的]为绵羊的生长发育研究提供基础资料。[方法]以2、30、60、90、120日龄雄性哈萨克羊和2、30-60、90日龄新疆细毛羊为试验动物,对其肌肉IGF-IR基因进行PCR扩增,并对扩增产物进行电泳检测和序列分析。[结果]绵羊肌肉IGF-IR基因PCR扩增产物在琼脂糖凝胶上出现了287和379bp2条带,该基因与牛IGF-IR基因序列的同源性为98.59%;在整个研究过程中,哈萨克羊1GF-IRmRNA表达量较低、较稳定,仅在30日龄时略有上升;新疆细毛羊IGF-IRmRNA表达量在2日龄时较高,30日龄时略有下降,60日龄时达到最高峰,90日龄时降到最低水平,且各日龄表达量差异显著;哈萨克羊mRNA表达量在2~60日龄时显著低于新疆细毛羊,而90日龄时显著高于新疆细毛羊。[结论]绵羊肌肉IGF-IR基因表达存在较大的品种间差异。[Objective] The study was to provide the basic data for studying growth and development of sheep. [Method] With kazak male sheep of 2, 30, 60, 90, 120 day-old and Xinjiang fine wool sheep of 2, 30, 60, 90 day-old as the tested animal, the IGF-IR gene in their muscle was amplified with PCR, and the amplified products was detected by electrophoresis and its sequence was analyzed. [Result] The PCR amplified products of IGF-IR gene appeared 2 strips with 287 and 379 by in agarose gel. The homology of IGF-IR gene in sheep and cow was 98.59% . During the whole research process, the expression level of IGF-IR mRNA in Kazakh sheep was lower and stable and slightly increased at 30 day-old. The expression level of IGF-IR mRNA in Xinjiang fine wool sheep was higher at 2 day-old, and slightly decreased at 30 day-old and reached the highest peak at 60 day-old, and decreased to the lowest level at 90 day-old, and the expression level at different. day-old was significantly different . The expression level of mRNA in Kazakh at 2 - 60 dayold was obviously lower than that in Xinjiang fine wool sheep, and was significantly higher than that in Xinjiang fine wool sheep at 90 day-old. [Conclusion] The IGF-IR gene expression in sheep muscle had big difference between varieties.
