摘  要:目的:探讨孕酮(P4)处理的树突状细胞(dendritic cell,DC)在诱导同种异基因移植免疫耐受中的作用。方法:实验小鼠分为4组,每组动物供体为C57BL/6雄性小鼠8只,受体为BALB/c雄性小鼠8只。体外培养供体小鼠骨髓来源DC,然后用P4处理。受体小鼠在皮肤移植前经尾静脉分别输注体外培养且经P4处理的供体小鼠DC、供体小鼠成熟DC以及不成熟DC,输注磷酸盐缓冲液为对照组。1周后受体小鼠进行同种异基因皮肤移植,手术后观察皮肤存活情况,应用流式细胞术检测手术前后受体小鼠外周血中CD4+CD25+调节性T细胞(CD4+CD25+T)百分率的变化。结果:在同种异基因皮肤移植中,受体小鼠尾静脉输注经P4处理的DC组分别与成熟DC组、不成熟DC组和对照组比较,移植皮肤存活时间显著延长(P<0.01);与成熟DC组、不成熟DC组和对照组比较,P4处理组外周血中CD4+CD25+T百分率显著升高(P<0.01)。结论:P4处理的DC可以延长同种异基因移植物存活时间。Objective: To investigate the effect of progesterone-treated dendritic cell (DC) on induction of allograft immune tolerance. Methods: Mice were divided into 4 groups, each group included 8 C57BL/6 donor and 8 Balb/c recipient mice. Bone marrow-derived DCs from C57BL/6 mice were cultured in vitro. Recipient mice received one injection of either progesterone-treated DC (P4 group), mature DC (mDC group) and immature DC (imDC group) through tail vein one week before transplantation, mice receiving phosphate-buffered saline served as the control. Skin transplantation was established and survival time of skin allograft was observed. The percentage of CD4+CD25+ T cell in peripheral blood was determined by flow cytometry before and after transplantation. Results: The survival time of the skin allograft was significantly prolonged in P4 group compared with that of mDC group, imDC group and the control (P〈0.01), accompanied with higher percentage of CD4+CD25+ regulatory T cell (P〈0.01). Conclusion: Dendritic cell treated with progesterone could prolong survival time of allograft.
