摘 要:[目的]筛选出适于工业化生产葛根素的提取、分离、纯化工艺路线。[方法]用熔点、红外光谱、核磁氢谱和紫外分光光度法分析鉴定提取的葛根素精品。[结果]葛根素精品的熔点为327~328℃,与葛根素标准品的熔点基本相同;在薄层色谱上,精品与标准品的斑点在紫外灯下均显蓝色荧光,Rf值均为0.31;精品的红外数据为IRUMax^Kbc(cm^-1):3398、1630、1584、1515、1443、1384、1257、1178、1118、1081、10141,标准品的红外数据为IRUmax^KBr(cm^-1):3370、1631、1607、1514、1447、1396、1273、1210、1103、1059、1008;精品的核磁氢谱数据与文献报道相符;在250nm处测定精品的吸光度,由回归方程计算出精品的葛根素含量为97%,液-液萃取法的葛根素精品的产率为1.41%。[结论]液-液萃取法分离纯化葛根素成本低,操作简单,可用于工业化生产。[Objective] The purpose of the study was to screen out the process route of extracting, isolating and purifying puerarin, which was suitable for industrial production. [Method] Tne extracted and refined puemrin was analyzed and identified by melting point method, infrared spectrophotometry,nuclear magnetic resonance hydrogen (NMR-H) spectroscopy and UV speetrophotometry. [ Result] The melting point of refmed puerarin was 327 - 328℃ and it was basically identical with the mehing point of standard puerarin. On thin layer spectrum, both the dots of refined and standard puerarin showed were IRUMax^Kbc(cm^-1) : 3 398, 1630, IRUMax^Kbc(cm^-1) : 3 398, 1630, blue fluorescence under UV-light lamp and both of their P,f values were 0.31. were IRUMax^Kbc(cm^-1) The infrared data of refmed puerarin KBr - : 3 398, 1 630, 1584, 1515, 1443, 1384, 1257, 1 178, 1 118, 1081andl041 and that of standard puerarin was IRUMax^Kbc(cm^-1)3 370, 1 631, 1 607, 1 514,1 447, 1 396, 1 273, 1 210, 1 103, 1 059 and 1 008.The NMR-H data of refined puerarin accorded with literature reports. The absorbance of refined puerarin was detected at 250 nm, the puerarin content of the refined was calculated to be 97% through regression equation.The yield rate of refined puerarin of liquid-liqiud extraction was 1.41 %. [Conclusion] Isolation and purification of puerarin by liquid-liqiud extraction was cheap in cost, simple to op-emte and could be used in industrial production.
