摘  要:[目的]探讨喜树碱含量最高的喜树叶片的最佳采摘时间。[方法]采用高效液相色谱法研究同一地区不同时间采摘的喜树嫩叶中的喜树碱含量差别。[结果]高效液相色谱的条件为:Kromasil C18柱(250 mm×4.6 mm,5μm),流动相为:甲醇∶水=6∶4(体积比),流速1.0 ml/min,检测波长为254 nm,柱温为室温,进样量20μl。样品制备条件:以甲醇为溶剂,60℃下超声提取1 h。高效液相色谱法测定喜树嫩叶中喜树碱最高含量为3.25‰。喜树嫩叶在4月中旬到4月末这段时间,喜树碱的含量是最高的,它比喜树果和喜树成叶都高,因此喜树嫩叶可作为喜树碱的生产原料。[结论]该研究为喜树碱的工业化生产奠定了基础。[ Objective] The aim was to discuss the optimum pick time of Camptotheca acuminate leaves with highest camptothecin content. [ Method] The difference of eamptothecin content from young leaves of C. acuminate picked in different tinge at the same area was studied by HPLC. [ Result ] The HPLC conditions were as follows: Kromasil C18 column (250 mm× 4.6 mm, 5μm), mobile phase was methanol water = 6:4 (volume ratio), flow rate was 1.0 ml/min, determining wavelength was 254 nm, column temperature was worn temperature and injection volume was 20 t.d. The sample preparation conditions were as follows: methanol was taken as solvent and ultrasonic extraction was taken for 1 h at 60 ℃ The highest content of camptothecin from young leaves of C. acuminate was 3.25%0, which was determined by HPLC. The camptothecin content of young leaves of C. acuminate was highest from the mid April to the end of April, which was higher than that from the fruit and mature leaves of C. acuminate. So young leaves of C. acuminate could be taken as materials for camptothecin production. [ Conclusion] The research laid the foundation for the industrial production of camptothecin.
