摘 要:[目的]筛选一套较为完善的适宜于水稻稻瘟病抗性叶片离体鉴定的方法。[方法]从保绿剂对离体叶保绿效果的影响、保绿剂对孢子在离体叶上萌发率的影响、不同孢子接种方法对叶片发病的影响和孢子悬浮液浓度、温度对稻瘟病病原菌染叶片效果的影响几方面对水稻稻瘟病抗性叶片离体鉴定方法的影响因素进行了研究。[结果]以6-BA作为保绿剂,浓度在50-75mg/ml范围内保绿效果较好,叶片几乎全部保绿。接种方法以喷雾法与液滴法较好,孢子萌发率较高,分别达到了84.7%、86.5%,浸泡法不适合叶片离体鉴定。温度为28℃时,接种用孢子悬浮液的浓度1×10^5~2×10^5个/ml较为合适。叶片发病期间温度低于28℃,潜育期有所延长。[结论]得到了行之有效的水稻抗瘟性叶片离体鉴定方法。[Objective] A set of complete identification method of the resistance of rice( Oryza sativa ) leaf in vitro to blast was screened. [Method] effect of the Green-maintaining Agent on the in vitro leaf color and the germination rate of spore and the effect of tire different inoculation methods on disease-forming in leaf, the effect of the concentration and temperature of spore suspension on rice leaf infected by blast pathogen were studied. [Results] The green color of all of leaves could be kept under the condition of the plant hormone-6-BA used as Green-maintaining Agent with the concentration of 50 - 75 mg/ml. The inoculation of spraying and dropping method promoted the high ratio of spore germination. The suitable concentration of the spore snspension was 1 × 10^5 - 2 × 10^5 spore/ml while the temperature was 28 ℃. The observation of symptom of infection on leaf would be delayed when the temperature was below 28 ℃. [Conclusion] The effective identification method of the resistance of rice leaf in vitro to blast was screened.
