摘 要:小泛素样修饰蛋白(SUMO)4是SUMO家族中新近发现的成员。SUMO的生化反应途径与泛素化有类似之处,但不同的是会使蛋白质更加稳定,而不是降解蛋白质。SUMO4可能是通过核因子(NF)-κB调控机制产生作用,拮抗泛素对NF-κB抑制蛋白(IκB)α的作用。而SUMO4 M55V变异可能导致SUMO化失效,泛素系统和NF-κB激活,介导免疫反应和炎性反应。并且已经得到证实的是,SUMO4基因M55V的多态性与某些人群的1型糖尿病相关。现在正在进行的一些研究试图揭示其在2型糖尿病发病机制中的作用。Small ubiquitin-related modifier (SUMO)4 is a newly found member of SUMO family. The biochemical effects of SUMO are somehow similar with those of ubiquitin, but SUMO stabilizes proteins, not degrades. The effect of SUMO4 is possibly mediated by NF-κB, with its antagonistic effect to ubiquitin. SUMO4 M55V mutation could repress sumoylation, activate ubiquitin and NF-κB,which leads to immunological responses and inflammation. It has already been verified that the polymorphism of M55V in SUMO4 is associated with type 1 diabetes in some populations. Some ongoing researches will try to explore its role in the pathogenesis of type 2 diabetes.
