摘 要:利用电子克隆方法获得葡萄乙醇脱氢酶基因Ⅲ(ADHⅢ),并采用生物信息学方法对该基因编码蛋白从氨基酸组成、理化性质、跨膜结构域、疏水性/亲水性、亚细胞定位、高级结构以及功能域等方面进行了预测和分析。结果表明,葡萄ADHⅢ基因全长1 602 bp,包含1 140 bp的ORF,编码379个氨基酸,该蛋白不具有明显的疏水区域,也无跨膜结构域,α-螺旋和不规则卷曲是其二级结构的主要构件。葡萄ADHⅢ包含有ADH功能域,和其他植物的ADHⅢ在序列组成、高级结构及活性位点等方面均具有高度的相似性。An alcohol dehydrogenase Ⅲ (ADHⅢ) gene from Vitis vinifera was cloned by electronic cloning based on the EST sequences from Unigene of NCBI. Some characters of the ADHⅢ that can encode amino acid were analyzed and predicted by the tools of bioinformaties in the following aspects,including the composition of amino acid sequences, hydrophobicity or hydrophilicity, secondary and tertiary structure of protein and function. Results showed that the full-length of ADH Ⅲ from Ⅴ. vinifera was 1 602 bp long and it contained a complete ORF which encoded 379 amino acid. The encoded protein had not obvious hydrophobicity region and transmembrane structure was not contained. The main motif of predicted secondary structure of ADH Ⅲ were alpha helix and random coil. The sequence, advanced structure and activity sites of ADHⅢ that contained an ADH domain were highly conserved compared with other plant ADH Ⅲ.
