摘 要:目的:研究不同月龄淀粉样前体蛋白(APP)转基因小鼠学习记忆功能,脑皮质,海马胆碱酯酶以及突触蛋白-Ⅰ的变化.方法:采用Morris水迷宫测试学习、记忆功能,比色法测定胆碱酯酶活性,免疫组织化学法测定突触蛋白-Ⅰ变化.结果:6,9,12mo APP阳性转基因小鼠(APP+)学习、记忆能力明显降低,9~12月龄降低尤为显著;皮层,海马胆碱酯酶活性降低,且皮层胆碱酯酶活性下降更为明显;海马CAⅠ区突触蛋白-Ⅰ的表达随着月龄的增加而减少.结论:APP转基因小鼠学习、记忆功能与海马内胆碱酯酶活性及突触蛋白-Ⅰ的表达呈负相关.说明在阿尔茨海默病(AD)的发病机制中乙酰胆碱的变化和神经突触的萎缩都是重要原因.AIM: To observe the changes of learning and memory ability, cholinesterase activity and synaptic proteins in hippocampus and cortex in transgcnie mice. METHODS: Morris water maze was used to study the learning and memory ability of action,colorimetry was used to detect the cholinesterase activity and synaptic proteins were observed by histochemistry. RESULTS: APP positive transgenic mice of different ages showed a decline in learning and memory, especially the age of 9 - 12 months. The cholinesterase activity in hippocampus and cortex greatly degraded and the degradation in cholinesterase activity of cortex was more severe. The quantity of synaptophysin- I decreased along with the increasing age in APP positive transgenic mice. CONCLUSION:The learning and memory ability is negatively correlated with synaptic proteins and the cholinesterase activity in APP transgenic mice.
