摘 要:本文利用随机扩增多态DNA技术研究了云南4个马品种48个个体的遗传变异和系统发育关系。在所使用的25个引物中,有22个引物扩增出多态谱带。共检测到216条扩增片段,其中74条出现变异。用Shannon指数计算了4个品种的遗传变异及其遗传变异在群体内和群体间的分布。用Nei的片段共享度公式计算了48个个体的遗传距离,用UPGMA聚类法构建了系统发育树状图。结果表明,腾冲马和新丽江马品种内的遗传变异显著高于文山马和乌蒙马,遗传变异68.06%存在于群体内。系统聚类图中品种内大多数个体聚在一起,文山马和乌蒙马聚成一个大类,腾冲马和新丽江马又聚成一个大类,说明文山马和乌蒙马以及腾冲马和新丽江马有较近的亲缘关系。In this study,random amplified polymorphic DNA(RAPD) analysis was used to estimate genetic diversity and relationship in 48 indivuduls which belong to four Yunnan horse breeds:Wenshan,Tengchong,Xin lijiang and Wumeng.25 primers were used,among which 22 generated polymorphic loci.A total of 216 bands were scored,of which 74(34.3%)were polymorphic.Shannon index was used to estimate the genetic diversity and the proportion of genetic variation within and between population.The genetic distance matrix was obtained by proportion of shared fragments.Dendrogram was generated by UPGMA.The results indicated that the genetic diversity of Tengchong and Xin lijiang was significantly higher than that of Wenshan and Wumeng.About sixty nine percert variation was detected within population.Most indivuduls in the same breeds were clustered together.Tengchong breed is closely related to Xin lijiang breed,and Wenshan breed is closely related to Wumeng breed.
