山西汉族人群DYS719基因座的遗传多态性研究

摘　　要：目的:调查基因座DYS719在山西汉族人群中的频率,并对其法医学意义进行探讨。方法:从山西汉族人群中随机抽取120例无关男性血样本,10例女性血样本,提取静脉血DNA,在Y染色体基因组DNA中查找候选的基因座,用GDB数据库中的引物进行PCR扩增,非变性聚丙烯酰胺凝胶电泳分离以及硝酸银染色,然后将筛选的等位基因进行测序,构建等位基因分型标准物,按照ISFG原则命名各等位基因。结果:共检出5个等位基因,其基因多样性为0.7384;法医学应用方面,其个人识别力和非父排除率均为0.7384。结论:Y染色体基因座DYS719具有较高的遗传多态性,在法医学及人类遗传学方面具有较高的应用价值。Objective：To research the genetic polymorphism of DYS719 gene loci in Shanxi Han population and provide data for Chinese Y-STR data base. Method：Venous blood samples were collected from 120 unrelated males and 10 females in Shangxi Han population.DYS719 gene loci was extracted from blood samples and amplified with PCR technique, then PCR products were analyzed by polyacrylamide gel electrophoresis and silver-stain.At last,gene diversity（GD）of DYS719 gene,the power of discrimination（PD）and the power of exclusion（PE）were calculated. Result：AAT repeat expansion of DYS719 gene loci has 11,12, 13,14,15 repeats respectively. Its GD was 0.7384,PD and PE in forensic science were 0.738 4. Conclusion：For human genetics and forensic genetics, the obtained data could be used to deal with the paternity test and individual identification.
