摘  要:目的:探讨养精种玉汤对KM小鼠骨髓细胞微核率的影响,对其遗传安全性进行研究。方法:采用不同时间点、不同剂量对KM小鼠进行体内骨髓细胞染毒,观察其致小鼠骨髓细胞染色体完整性受损及染色体分离异常等情况。结果:采用不同时间点取样,其骨髓细胞微核率与溶剂对照相比无明显增加(P>0.05),说明在时效关系上,不同时间点给药对小鼠骨髓细胞微核率无明显影响。采用不同剂量进一步染毒,其骨髓细胞微核率分别与正常对照相比也无明显增加(P>0.05),说明在量效关系上,无明显诱发小鼠骨髓细胞微核率增加作用。正常对照组和高剂量组嗜多染红细胞与成熟红细胞比值未见明显降低,表明该药对小鼠骨髓细胞也无明显毒性。结论:养精种玉汤药液100 g/kg的剂量范围以内,初步认为无明显致小鼠骨髓细胞染色体完整性受损及染色体分离异常。Objective: To investigate the effect of Yangjing Zhongyu Decoction on marrow cell micronucleus rates in KM mice, and study its genetoxic. Method: The mice were poisoned in bone marrow at the different time points, and with different doses of Yanking Zhongyu Decoction, we probed into mammalian marrow cell micronucleus rates for Yangjing Zhongyu Decoction, and observe the damage on integrality of bone marrow cell and abnormality of chromosomal disjunction. Result: When we sampled in different time points, marrow cell micronucleus rates had not been raised contrasting to the solvent (P 〉 0.05 ). That was to say, administration in different time points had no effect on marrow cell micronucleus rates in time - effect relationship. Poisoning in bone marrow with different dosages could not raise up marrow cell micronucleus rates also ( P 〉 0.05 ), that was to say, in dose - effect relationship, dosage couldnt induce marrow cell micronucleus rates increasing. In solvent control group and high dose group, that the decoction could not decreasing polychromatic erythrocyte erythrocyte and mature erythrocyte markedly showed that the decoction had no overt toxic effect. Conclusion: Yanking Zhongyu Decoction has no the effect of damage of bone marrow cell integrality and abnormality of chromosomal disjunction in the dosage range of 100 g/kg.
