摘 要:目的:探讨降脂益肝冲剂对高脂饮食诱导的大鼠非酒精性脂肪性肝炎过氧化物酶体增殖物活化受体γ(PPARγ)的影响。方法:40只雄性Wistar大鼠随机分为4组(每组10只):正常组给予普通饲料喂养,模型组和治疗组给予高脂饮食喂养。治疗组在高脂饮食8周后给予降脂益肝冲剂,同时模型组和正常组分别给予等量的生活饮用水灌胃,12周末处死实验大鼠。测定空腹血糖(FBG)、空腹胰岛素(FINS),计算胰岛素敏感指数(ISI);测定血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆固醇(TC)、甘油三酯(TG);HE染色观察肝组织病理变化;RT-PCR分别检测大鼠肝脏PPARγ mRNA的表达。结果:模型组大鼠血清转氨酶、TC、TG升高,ISI降低,伴典型的脂肪性肝炎;与模型组相比,治疗组的各项指标都有所改善,且两组间有统计学差异。模型组大鼠肝组织PPARγ mRNA表达减少,降脂益肝冲剂能增加脂肪肝大鼠肝组织PPARγ mRNA的表达。结论:降脂益肝冲剂能通过调节PPARγ的表达来调节脂质代谢,改善胰岛素抵抗,降低肝脏的脂质沉积,有效地治疗大鼠非酒精性脂肪性肝炎。Objective: To observe the effect of Grannul ofJiangzhi Yigan on peroxisome proliferator-activated receptors γ ( PPAR - γ) in the fatty liver tissue of rats with nonalcoholic fatty hepatitis chronically induced by high fat diet. Method: 40 male Wistar rats were randomly divided into 4 groups (10 rats in each group). The rats in normal group were fed up with standard diet, the ones in model group and treatment group were fed up with high -fat diet. After 8 weeks, Grannul of Jiangzhi Yigan was given to treatment group by intragastric administration, and life drinking water was given to normal group and model group. After 12 weeks, all the rats were put to death. The level of aminotransferase (ALT and AST), total cholesterol (TC) and triglyeride (TG) were detected by automatic biochemistry analyzer, fasting blood glucose (FBG) was detected by glucose oxidase method, serum insulin was detected by radio -immunity assay, and the insulin sensitivity index(ISI) simultaneously was calculated. The pathological changes of hepatic tissue were observed by microscopy after stained by hematoxylin and eosin (HE). Expression of PPARγ in the liver was detected by reverse transcriptase - polymerase chain reaction ( RT - PCR). Result: ALT, AST, TC and TG in model group were developed, but ISI was decreased companying with fatty hepatitis. Compared with model group, every data in treatment group improved. There was statistical difference between the two groups. Expression of PPARγ mRNA in liver tissue in model group was decreased. Grannul of Jiangzhi Yigan could increase the expression of PPARγ mRNA in liver tissue of rats. Conclusion: Grannul of Jiangzhi Yigan can heal nonalcoholic fatty hepatitis of rat effectively through regulating PPARγ to regulate lipid metabolism and improve insulin resistance to depress lipidoses in liver.
