摘  要:目的探讨人血管生成素1(Ang-1)对人胃癌细胞BGC-823基质金属蛋白酶2(MMP-2)表达情况的影响。方法利用腺病毒作为载体将已构建成功的含Ang-1基因的重组腺病毒(Ad-Ang-1)瞬时转染人胃癌细胞BGC-823为实验组,转染编码绿色荧光蛋白的复制缺陷型腺病毒重组体(Ad-GFP)为阴性对照组,未转染的正常细胞为对照组。应用RT-PCR和Western blot印迹检测三组细胞中MMP-2在mRNA和蛋白表达的情况。结果MMP-2在实验组中表达明显高于阴性对照组和空白对照组(P<0.05)。结论Ang-1能显著提高人胃癌细胞BGC-823的MMP-2的表达,提示Ang-1可能具有促进肿瘤细胞侵袭的作用。Objective To explore the effects of angiopoietin-1 on matrix metalloproteinase-2 (MMP-2) of human gastric cancer cell line BGC-823. Methods The cell line of BGC-823 was transfected with Ad-Ang-1 and Ad-GFP. The cell line of BGC-823 without transfection was taken as the control. The expression of MMP-2 mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). The protein expression of MMP-2 was detected by Western blot. Results The expression of MMP-2 was significantly higher in Ad-Ang-1 transfected group than that in transfected group and Ad-GFP group (P〈0.05). Conclusion Transfection of Ang-1 into human gastric cancer cell line BGC-823 can significantly increase the expression of MMP-2 suggesting that Ang-1 may promote tumor invasion.
