寻常性银屑病患者外周血CD14^＋单核细胞TLR4的表达及其与TNF-α，IL-6的相关性

摘　　要：目的探讨寻常性银屑病患者外周血单核细胞Toll样受体4(Toll-like receptor4,TLR4)的表达及其与TNF-α,IL-6浓度的相关性,初步探讨TLR4在寻常性银屑病发病中的作用。方法采用流式细胞术检测30例寻常性银屑病患者外周血CD14+单核细胞TLR4的表达,并采用双抗体夹心酶联免疫吸附法(ELISA)检测血清TNF-α,IL-6的浓度。另取20例正常人作为对照组。结果寻常性银屑病患者外周血中单核细胞TLR4的表达、血清TNF-α,IL-6浓度较正常人对照组明显增高(P<0.01),TLR4表达与血清炎症因子TNF-α,IL-6浓度变化呈正相关(P<0.05)。结论TLR4及其介导的天然免疫应答在银屑病发病机制中起重要作用,其机制可能是通过上调TLR4的表达促使炎症因子产生和分泌而介导银屑病的发病。Objective To explore the expression of TLR4 on human peripheral blood mononuclear cell and its correlation with concentration of TNF-α and IL-6 in the blood of patients with psoriasis vulgaris. Methods The positive rates of TLR4 in human PBMC were measured by flow cytometry in 30 cases of patients with psoriasis vulgaris and normal controls. Levels of TNF-α and IL-6 in sera were measured by double antibody sandwich enzyme-linked immunosorbent assay （ELISA）. Results The positive rates of TLR4 in human leucocytes and the serum levels of TNF-α and IL-6 were significant increase in patients with psoriasis vulgaris as compared with that of normal controls （ P 〈 0.01 ） , and the expression of TLR4 was positively correlated with the concentration of TNF-α and IL-6. Conclusions The TLR4 and the innate immune response mediated by it may play a critical role in the pathogenesis of psoriasis vulgaris, which may be through spurring the excretion of inflammatory factor.
