摘  要:目的:研究首乌仙海片激活下调脑区治疗缺血性卒中的作用机制。方法:电凝法制备MCAO大鼠模型。180只大鼠随机分为模型组、药物组和假手术组。于造模成功后药物组按4.5g生药/kg灌胃给予首乌仙海片,余二组分别灌胃给予同等量生理盐水,日1次,共2周。以BWT评分法进行神经行为学评价,结合局部脑血流量(rCBF)测定,免疫组化和原位杂交法分别观察大鼠脑组织生长相关蛋白(GAP-43)、突触素P38(SYN)的表达和大鼠脑微血管内皮细胞ET-1、ecNOS、P-gp mRNA的变化。结果:模型组大鼠病灶远隔区存在低灌注现象,突触活性降低,脑微血管内皮细胞对脑微循环的调节机能下降;药物组可明显改善这种低活动状态,并能显著提高大鼠运动功能。结论:首乌仙海片对缺血性卒中模型大鼠神经功能有显著改善作用,激活下调脑区是其重要机制。Objective:To study the action mechanism of shouwuxianhai tablets ischemic cerebrovascular disease from the activation cerebral area of functional down-regulation. Methods: 180 rats were divided into model group,drug group and pseudo-operation group at random. The rat models of MCAO were successfully made by electrocoagulation, the rats of drug group had been taken orally shouwuxianhai tablets by 4.5g/kg crude drug one time daily for two weeks, the rest groups had been taken orally equivalence Saline one time daily for two weeks. Neuroethology were evaluated with BWT scoring standard, and regional cerebral blood flow (rCBF)were measured, the expresses of GAP-43.SYN in the brain tissues of experimental rats and the change ofET-1, ecNOS. P-gp mRNA in the brain microvessel endothelial cell of experimental rats were observed by means of immunohistochemical method and hybridization in situ. Results The focus remoteness areas of model rats presented hypoperfusion phenomena. The synapse activity and the function of brain microvessel endothelial cell accommodating brain microcirculation descended in the model group. Hoverer, the low activity states were obviously improved in the drug group, and the traditional Chinese drug could notably enhance the motor function of experimental rats. Conclusions: The shouwuxianhai tablets can obviously improve the nerve function of model rat of ischemic cerebrovascular disease. The activation of cerebral area of functional down-regulation is important mechanism of shouwuxianhai tablets action.
