摘 要:为了研究CD47分子在猪附红细胞体病溶血中的作用,本研究利用去脾猪人工诱导了猪附红细胞体病。在试验猪发病的不同时期采集血样,对各期血样进行了红细胞计数,并用FACScan流式细胞仪在480 nm荧光下检测了红细胞表面CD47分子的表达量。红细胞计数结果显示:第7 d轻度变形的红细胞明显增多;第10 d几乎看不到正常红细胞,轻度变形和严重变形红细胞占优势;第28 d随病情的恢复血液中几乎都是正常的红细胞,红细胞异型程度虽病情发展而变化。流式细胞术结果显示:阳性细胞荧光强度百分比(M1)的最低值发生在接种后的第7 d,比对照组低82%,是发病症状的高峰期。第14 d和第21 d测得的M1值渐渐升高,但是仍显著低于对照组。这证明红细胞上CD47分子表达量与红细胞异型程度变化相一致。由于红细胞上CD47分子数量的减少减弱了CD47-SIRPα的交互作用,从而为脾脏红髓内巨噬细胞吞噬红细胞提供了正性信号。实验结果表明,作为红细胞表面标记物的CD47分子在猪附红细胞体病溶血机制中发挥着重要作用。To evaluate the alteration of CD47 on red blood cells (RBCs) in pigs infected with M.suis, porcine eperythrozoonosis (PE) was experimentally induced and blood samples collected at the different time points. The results showed that the change in erythrocyte count is closely related with the development of illness. Abnormal erythrocytes started to increase at 7 days post-inoculation, and majority of RBCs became abnormal in shape at 10 days post-inoculation. Almost no erythrocytes existed at 28 days postinoculation. Flow cytometry analysis indicated that the CD47 count on RBCs changed correlatively with the course of PE. The lowest value of M1 (percentage of the positive cells in fluorescence intensity), which was 82 % lower than the control sample, occurred on day 7 post-inoculation at the peak of parasitemia. The M1 value increased slowly on day 14 and 21, but still significantly lower than the controls. These results suggested that the number of CD47 on RBCs altered progressively with the phases of the PE disease. The decrease of CD47 on RBCs may weaken the inhibitory CD47-SIRPα interaction and provide positive signals for phagocytosis of macrophages resulting in the removal of the RBCs from the circulation. Therefore, CD47, a Marker on RBCs, may play an important role on the mechanism of hemolysis caused by infection of M.suis.
