摘 要:重组蛋白质技术作为蛋白质研究的重要手段之一,在生物化学与生物物理学研究领域,扮演着极其重要的角色。亲和纯化作为最为方便与快捷的重组蛋白质纯化手段,日益得到广泛的应用。由于各种亲和标签,纯化介质层出不穷,性质各异。应根据自身研究对象具体情况选择合适的亲和纯化标签。近年双亲和标签进行串联亲和纯化日益成为蛋白质相互作用研究的重要方法。多种亲和标签的搭配已得到成功应用,部分已进入商业化,并在各种模式生物中得到广泛的应用,本文就重组蛋白质亲和标签的选择与串联亲和纯化作一综述。As a key method of protein study, the recombinant protein technology is playing an essential role in biochemical and biophysical study. The affinity purification is widely used as the most convenient method of protein purification. There are several kinds of affinity tags and purification resin in application with different characters. The choosing of the appropriate affinity purification strategy should be guided according to the specific character of the study object. With two affinity tags used in order, researchers can purify protein complex with high purity. This strategy, which is also named tandem affinity purification, is now an important method used in protein interaction discovery. Several kinds of tag matching are successfully used in previous study and some have been successfully commercialized. The tandem affinity purification has now been widely applied in all kinds of model organisms.
