摘 要:目的探讨rh-bFGF治疗下肢缺血的效果及最佳剂量。方法30只实验兔,建立下肢缺血模型。随机分为5组,A组为对照组,B、C、D、E组为治疗组,分别经肌肉局部注射rh-bFGF2.5μg,5μg,10μg,20μg。行血管造影、毛细血管密度、微血管/肌纤维束比值及病理学检查。结果1.下肢动脉造影:D、E组侧支血管数均多于对照组及B、C组(P<0.05)。2.微血管密度和微血管/肌纤维束比值:D、E组较对照组和B、C组增高(P<0.05)。3.病理学观察:D、E组血管生成较A、B、C组多。结论应用实验剂量rh-bFGF治疗肢体缺血是安全有效的;其疗效随着rh-bFGF剂量的增加而增强,rh-bFGF治疗肢体缺血的最佳剂量为10μg。Objective To study the therapeutic effect and optimal dose of recombinant human basic fibroblast factor (rh-bFGF) to treat low limb ischemia. Methods 30 rabbits were used to establish the animal model of lower limb ischemia. 5 groups were randomly separated. A group was control one; B, C, D and E groups were treatment groups, and 2.51μg, 5μg, 10μg, 20μg of bFGF were respectively administered by the intramuscular injection. Digital subtraction angiography of lower limb, microvessel density and microvessel density/muscle fiber ratio were examined Results 1. Digital subtraction angiography of lower limb: The number of collateral vascular in D and E groups was higher than A, B and C groups (P〈0.05).2. Microvessel density and microvessel density/muscle fiber ratio: D and E groups were higher than A, B and C group (P〈0.05). 4. Pathological examination: The angiogenesis of D and E groups was more than A, B and C groups. Conclusion The experimental dose ofbFGF to treat lower limb ischemia is safe and effective. The curative effect is better with the dose of bFGF increaces. The best dose of bFGF to treat lower limb ischemia is 10μg.
