摘  要:[目的]为改良渤海黑牛地方品种及种质资源的保护提供技术依据。[方法]根据GenBank发表的牦牛心脏脂肪酸结合蛋白基因(H-FABP)的序列设计1对引物,利用PCR技术扩增出渤海黑牛H-FABP基因外显子2的DNA片段,测定其核苷酸序列并进行氨基酸序列的推导,同时将序列与牦牛、山羊、猪、人、小鼠和鸡的外显子2序列进行比较,并且进行系统发生树的构建。[结果]渤海黑牛的H-FABP外显子2核苷酸序列和氨基酸序列与牦牛、山羊、猪、人、小鼠和鸡各物种间同源性分别为100%、97.1%、94.2%、89.0%、83.8%、79.8%,100%、98.2%、94.7%、86.0%、86.0%、77.2%。系统发生结果显示,系统发生树总体分为两支,鸡单独为一支;渤海黑牛、牦牛、山羊、猪、人、小鼠聚在一起成为另一支。[结论]渤海黑牛H-FABP外显子2具有很强的保守性,H-FABP基因外显子2进化树符合物种进化规律。[Objective] The study aims to provide the technical basis for improving local variety of Bohai Black Cattle and resource protection.[Method] According to Genbank combined with the H-FABP gene,sequences of yak,a pair of primers are designed.The Exon 2 of Heart Fatty Acid-Binding Protein(H-FABP) gene of Bohai Black Cattle is amplified and sequenced directly.Then the acquired sequence is aligned to further identify it and deduce the amino acid sequence with the homologous Exon 2 of H-FABP gene of other species.A phylogenetic tree of the exon 2 of H-FABP gene of Bohai Black Cattle and other species is constructed with MEGA 4.0.[Result] After comparison,the identities of the nucleotide sequence and amino acid sequence of exon 2 of Bohai Black Cattle H-FABP gene with that of yak,goat,pig,human,rat and chicken are 100%,97.1%,94.2%,89.0%,83.8%,79.8% and 100%,98.2%,94.7%,86.0%,86.0% and 77.2%,respectively.The phylogenetic result shows that these species are classified into 2 branches as a whole,in which the chicken is on one branch and the Bohai black cattle,yak,goat,pig,human and mouse is on the other branch.[Conclusion] The exon 2 of H-FABP gene of Bohai black cattle is highly conserved.The phylogenetic tree of the exon 2 of H-FABP gene accords with the order of evolution.
