摘 要:目的观察月见草油和维生素E对D-半乳糖致衰老小鼠骨丢失的预防作用,从骨代谢方面探讨其可能的机制。方法40只8周龄昆明雌性小鼠随机分为4组:青年对照组,衰老模型组,维生素E组,月见草油组,每组10只,以颈背部皮下注射半乳糖1000mg·kg-1·d-1造成亚急性衰老模型,并预防灌胃用药6w,观察月见草油和维生素E对小鼠骨丢失时骨量、骨长度、骨有机质、骨钙、磷等骨矿物质以及血清骨代谢相关酶活性的影响。结果D-半乳糖可使小鼠骨量、骨长度、骨有机质减少(P<0.05),血清碱性磷酸酶(AKP)活性下降(P<0.01);同时给药月见草油和维生素E分别能使骨量、骨长度、骨矿物质及骨有机质得到恢复(P<0.05或P<0.01)并提高血清AKP活性(P<0.01),月见草油还能有效抑制抗酒石酸酸性磷酸酶(StrACP)活性(P<0.05)。结论D-半乳糖可导致小鼠骨丢失,月见草油和维生素E均可预防衰老模型小鼠骨丢失,而其机制可能与成骨细胞、破骨细胞活性的调控有关。Objective To study the protective effects of evening primrose oil(EPo) and Vitamin E(Vit E)on osseous changes in the aging mice induced by D-galactose (D-gal) and their mechanisms of bone metabolism. Methods 40 female Kunming mice were injected with D-gal for 6 weeks to establish the aging models. Bone routine parameters such as bone dry weight, bone length, bone hydroxyproline content, bone calcium, bone microelements and bone metabolism parameters such as serum alkaline phosphatase (AKP) activity, serum tartrate-resistant acid phosphatase (StrACP) activity were tested and compared among control, D-gal (model), Vit E and EPo groups. Results The bone dry weight, bone length, bone hydroxyproline content and serum AKP activity were significantly decreased in model group than those in control group(P〈0.05 or P〈0.01). The above changes of Vit E and EPo groups were significantly higher than those in model group (P〈0.05 or P〈0.01). Vit E and EPo also enhanced the content of bone calcium and microelements (P〈0.05 or P〈0.01). However, EPo decreased serum StrACP activity (P〈0.05) comparing with that in control group. Conclusions The high does of D-gal for short term could induce aging related osteopenia. EPo and Vit E could improve osseous changes induced by D-gal. These changes probably are related to the regulatory effects of the activities of osteoblast and osteoclast.
