摘 要:目的:观察阿托伐他汀短期干预对血脂异常兔血清及脂肪组织分泌脂肪细胞型脂肪酸结合蛋白(AFABP)的影响,并阐明其可能机制。方法:15只新西兰大白兔随机均分为3组:①对照组,给予普通饮食12周;②高胆固醇组,给予高胆固醇饲料12周;③阿托伐他汀组,在饲以高胆固醇饲料8周的基础上给予阿托伐他汀1.5mg·kg-1.d-14周。应用RT-PCR技术测定脂肪组织AFABP mRNA的表达;ELISA法检测血清及脂肪组织培养液中AFABP水平。结果:实验第8周、第12周时高胆固醇组血清总胆固醇、低密度脂蛋白胆固醇水平明显高于对照组(P<0.01);阿托伐他汀组经阿托伐他汀干预后对血脂产生明显影响,并能降低血清及脂肪组织上清液AFABP水平,与高胆固醇组比较,均P<0.05。阿托伐他汀降低血清AFABP水平与降低脂肪组织上清液AFABP水平呈正相关(r=0.943,P<0.05)。RT-PCR结果显示,高胆固醇组脂肪组织AFABP mRNA表达高于对照组(P<0.05);阿托伐他汀组较高胆固醇组能降低兔脂肪组织AFABP mRNA的表达(P<0.05)。结论:阿托伐他汀能降低血脂异常兔血清及脂肪组织上清液AFABP水平,这可能是阿托伐他汀抗动脉粥样硬化的多效性机制之一。Objective: To observe the effects of atorvastatin on serum adipose fatty acid-binding protein (AFABP) concentration and AFABP secretion by adipose tissue from dyslipidaemia rabbits. Method:Fifteen male New Zealand white rabbits were randomly divided into three groups: ①control group: fed with normal diet for 12 weeks;②high cholesterol group., fed with high cholesterol diet for 12 weeks; ③atorvastatin group: same cholesterol diet for 8 weeks supplemented with atorvastatin (1.5 mg · kg^-1· d^-1) for 4 weeks. AFABP concentrations in serum and adipose tissue culture supernatant were measured by ELISA. Adipose tissue were harvested for RTPCR studies. AFABP concentrations in serum and adipose tissue culture supernatant were measured by ELISA. Result:Rabbits fed with high cholesterol diet showed higher serum levels of total cholesterol, low density lipoprotein cholesterol than those fed with normal diet (P〈0. 01). Atorvastatin treatment can change serum lipids levels during the feeding period, and decreased AFABP concentration in serum and adipose tissue cultured supernatant in atorvastatin group compared with high cholesterol group (P〈0.05). The mRNA levels of AFABP in adipose tissue was increased in high cholesterol group, atorvastatin decreased the mRNA levels of AFABP in adipose tissue. There existed significantly positive correlation between decreases in serum AFABP value and subcutaneous adipose tissue cultured supernatant AFABP value in atorvastatin group (r=0. 943, P〈0.05). Conclusion:Our study indicated that atorvastatin reduced AFABP concentration in serum and adipose tissue culture supernatant of dyslipidaemia rabbits. These effects might be of another pleiotropic direct mechanism for the antiatherosclerotic actions of atorvastatin.
