摘  要:目的初步探讨Wnt10b在小鼠毛囊生长周期中的表达变化规律及其意义。方法首先用RT-PCR检测Wnt10b在小鼠皮肤中的表达情况,然后以免疫组化检测Wnt10b在毛囊生长周期各阶段的表达定位及变化,同时检测了核增殖抗原Ki67在毛母质中的表达情况。结果Wnt10b在生后第1周期生长期皮肤的mRNA表达量显著高于退化期和静止期(P<0.01),生长中期达到峰值,退化期和静止期呈低表达;第2周期生长早期表达量略高于退化期和静止期。Wnt10b蛋白特异性表达于生长期毛囊的毛母质(Matrix)部位;退化期和静止期在整个毛囊都不表达;第2周期生长早期主要表达于毛囊次级毛芽。Ki67在毛母质中的表达变化情况与Wnt10b具有一致性。结论Wnt10b的表达具有严格的时空特异性,生后第1周期特异性表达于生长期毛囊的毛母质细胞,第2周期生长早期表达于毛母质细胞的祖细胞次级毛芽,提示Wnt10b可能对毛母质细胞增殖起作用,进而调控毛囊的生长并维持毛囊周期。Objective To detect the expression pattern of Wnt10b in the hair follicles of mice at different age and explore its importance. Methods The expression of Wnt10b in the back skin of mice at anagen phase (1, 7 and 15 d after birth), catagen phase (18 d), telogen (23 d) and secondary anagen phase (29 d) was detected by reverse transcription-polymerase chain reaction (RT-PCR). The localization of Wnt10b in the hair follicle of same skin area of mice was measured by immunohistochemical method. The expression of Ki67 was studied at the same time. Results Wnt10b mRNA was strongly expressed in the skin at anagen phase (P 〈 0.01 ) , reached its maximum at the middle of anagen phase, and slightly expressed at catagen and telogen phases. In the secondary cycle, Wnt10b transcripts level in the early anagen phase was a little bit higher than at catagen and tclogen phases. Wnt10b protein was specifically expression in the hair matrix of anagen follicles. No such protein was detected at catagen and telogen follicles. The expression of Wntl0b appeared in the sec- ondary hair germ at early anagen in secondary cycle. Conclusion The expression of Wnt10b is strictly special in time and space. Wnt10b is specifically localized in the hair matrix at anagen, and in the secondary hair germ at early anagen in the secondary cycle. Accordingly, our results indicate that Wnt10b may promote the proliferation of matrix cells, and regulate the cyclical growth of hair cycles.
