摘  要:目的:观察急性有氧运动对PPARδ表达和相关代谢酶活性的影响,以研讨耐力训练使骨骼肌有氧代谢能力产生适应性变化的机制。方法:6周龄雄性C57BL/6小鼠,体重为15±2g,对照组8只,运动组32只。运动组以20m/min速度在跑台上运动120min,并在运动后的即刻、1h、3h和6h分别处死8只小鼠。采用RT-PCR和Western blot方法检测骨骼肌中的PPARδ、CPT-1和BoAcDH的表达。结果:与对照组相比,运动组PPARδ的mRNA和蛋白表达量分别增加了2.3倍和1.0倍(P<0.05),CPT-1和BoAcDH的蛋白表达分别提高了1.3倍和1.9倍。结论:急性有氧运动诱导了PPARδ的过量表达,从而调控脂肪酸氧化相关酶的活性,促进骨骼肌有氧代谢能力增强,成为长期训练使肌肉产生适应性变化的基础。Objective: Observing the influence of acute aerobic exercise on the expression of PPARδ and activity of some interrelated enzymes in order to study the mechanism of aerobics causing skeletal muscle biologically adaptive change. Method: male mice (C57BL/6), 6-week- old, 15±2g in weight, 8 in control group and 32 in exercise group. The mice in exercise group run 120mins at a speed of 20m/min, and they were killed immediately, an hour, three hours and six hours after exercises respectively. RT-PCR and Western blot are applied to examine the expression of PPARδ, CPT-1 and BoAcDH in skeletal muscles. Results: In comparison with control group, the expression of mRNA and protein in PPARδ has increased respectively by 2. 3 times and 1 time(P〈0.05)in the exercise group, and the expression of protein in CPT- land BoAcDH has increased respectively by 1.3 and 1.9 times. Conclusion: Acute aerobic exercise induced the excessive expression of PPARδ, thus adjusted the activity of interrelated enzyme in oxidizing fatty acid and enhanced the increase in the ability of aerobic metabolism to become the change basis of adaptability caused by long-term training.
