摘 要:双生病毒是全球性的重要植物病毒,为提高植物对此病毒的抗性,分离了烟粉虱体内GroEL基因,将其连接到棉花韧皮部特异启动子PGHNBS下游,构建到植物表达载体PBI121上,采用农杆菌介导法转化烟草,获得抗性苗。对抗性植株进行PCR、RT-PCR和病毒侵染检测,结果表明:GroEL基因已经整合到烟草基因组中,并且在转录水平上表达。病毒侵染检测结果显示转基因烟草植株中没有检测到病毒,证明韧皮部特异表达GroEL基因抗双生病毒是可行的。The geminivirus is a devastating virus all over the word. In order to develop an effective approach to control this virus, the GroEL gene from Bemisia tabaci which was cloned into PBI121 and drived under phloem specific promoter PGHNBS was isolated. Tobacco plants were transformed by co-cultivating leaves method via Agrobacterium mediation. The regenerated kanamycin resistant tobacco plants were confirmed by PCR, RT-PCR and identification of virus infection. PCR and RT-PCR proved the stable integration of the GroEL gene in tobacco genome and expression in transcription level. Virus infection experiment demonstrated geminivirus was not detected in transgenic plants. These results proved that phloem specific expression of GroEL gene has a good effect against geminivirus.
