摘 要:目的:探讨治疗型关节炎护膝防治膝骨性关节炎的作用机制。方法:日本大耳白兔54只,采用改良Hulth法复制膝骨性关节炎模型,随机分为6组,分别测定血液及关节滑膜MDA、SOD、NO浓度。结果:模型组、对照组、实验1组、实验2组、实验3组血液MDA、NO浓度均高表达,含量16周明显高于8周时,而血液SOD浓度均低表达,含量16周明显低于8周时,与正常组比较差异均有统计学意义(P<0.05)。8周时,血液中MDA、NO、SOD含量,对照组、实验1组、实验2组、实验3组与模型组比较差异均有统计学意义(P<0.05);对照组、实验1组、实验2组与实验3组比较差异均有统计学意义(P<0.05)。16周时,血液和关节滑膜中MDA、NO、SOD含量,对照组、实验1组、实验2组、实验3组与模型组比较差异均有统计学意义(P<0.05);对照组、实验1组、实验2组与实验3组比较差异均有统计学意义(P<0.05)。16周时与8周时,血液中MDA、NO、SOD含量,实验3组间比较差异均有统计学意义(P<0.05)。结论:OA护膝通过有效抑制氧自由基、NO的生物学效应、增强SOD的生物学活性来延缓关节软骨的退变。Objective: To investigate the mechanism of osteoarthritis (OA) kneepad on prevention and treatment of knee OA. Methods:Fifty--four Japan rabbits were randomly divided into 6 groups. Knee OA models were made by the method of modified Hulth. MDA, SOD and NO concentrations in both blood and synovial fluid were measured respectively. Resuits:High concentrations of blood MDA and NO were observed in model group, control group and 3 experimental groups. The concentrations were obviously higher in the 16th week than in the 8th week. Meanwhile, SOD concentration was low, with lower concentration in the 16th week than in the 8th week. Compared with control group, MDA and NO in synovial fluid significantly increased and SOD in synovial fluid statistically decreased (P〈0.05) at 16th week. At the 8th week, there were significant differences of serum MDA, NO and SOD between control group, 3 experimental groups and model group (P〈0.05). Meanwhile, significant differences of those indexes were found between control group and 3 experimental groups (P〈0.05). At the 16th week, there were significant differences of MDA, NO and SOD in serum and synovial fluid between control group, experimental groups and model group (P〈0.05). And significant difference of those indexes was found between control group and 3 experimental groups (P〈0.05). At the 16th and 8th week, there was significant difference of serum MDA, NO and SOD between the 3 experimental groups (P〈0.05). Conclusion:OA kneepad is effective to postpone degenerative change of arthrodial cartilage via inhibiting the biological effect of oxygen free radical and NO and promoting the biological effect of SOD.
