阴证、阳证皮肤溃疡创面渗出液MMPs和TIMPs的酶活性对比分析

摘　　要：目的:探讨皮肤溃疡阴证、阳证创面渗液基质金属蛋白酶及组织抑制因子活性和水平的差异,揭示其证候基础。方法:收集皮肤溃疡阳证20例、阴证25例创面渗液及10例健康成人血清对照,通过明胶酶谱法检测MMP-2、MMP-9的活性和ELISA法检测MMP-2、MMP-9、TIMP-1、TIMP-2的含量。结果:阴证创面MMP-2、MMP-9活性和含量不同程度高于阳证组和对照组(分别P<0.01);同时其TIMP-1、-2含量却不同程度低于阳证组和对照组(分别P<0.01),且MMP-9/TIMP-1、MMP-2/TIMP-2比值明显高于阳证组和对照组(分别P<0.05)。结论:阴证疮疡MMPs/TIMPs平衡失调,这可能是皮肤溃疡创面不易愈合的机制之一。Objective： Try to study the characteristic of traditional Chinese medicine （TCM） differentiations of chronic wound by analysis of matrix-metalloproteinases and their inhibitors in the skin wound fluids from ulcers of Yin and Yang syndromes. Methods ： 20 patients with skin ulcers of Yang syndrome （Yang group） ,25 patients with skin ulcers of Yin syndrome （Yin group） andl0 normal people（Control group）were studied, wound fluid activity of MMP-2 and MMP-9 were measured by gelatin zymogram, wound fluid levels of MMP-2,MMP-9 and TIMP-1,TIMP-2 were measured by enzyme linked immunoassay. Results： The activity and levels of MMP-2, MMP-9 were increased in skin ulcers of Yin syndrome compared with Yang group and Control group（ P 〈 0.01）,but the TIMP-1,TIMP-2 levels in Yin group was significantly lower than that in Yang group and Control group（ P 〈 0.01）. The ratio of MMP-9/TIMP-1, MMP-2/TIMP-2 was significantly higher in Yin group than that in Yang group and Control group（ P 〈 0.05） . Conclusion： Unbalance of MMPs/ TIMPs is a possible important mechanism of the delaying wound healing for skin ulcer with Yin syndrome.
