摘  要:【目的】观察石菖蒲有效成分β-细辛醚对缺血—再灌注损伤(MI-RI)心肌细胞的保护作用。【方法】培养原代乳鼠心肌细胞,采用连二亚硫酸钠(Na2S2O4)复制心肌细胞MI-RI模型,以四甲基偶氮唑盐(MTT)法测定细胞存活率,紫外分光光度法测定细胞培养液中乳酸脱氢酶(LDH)和肌酸激酶(CK)的含量,利用流式细胞术(FCM)检测线粒体膜电位(MMP)平均荧光强度。【结果】β-细辛醚能显著提高MI-RI模型细胞存活率,降低培养液中LDH、CK的含量,提高MMP值(均P<0.05)。【结论】β-细辛醚对MI-RI心肌细胞具有保护作用,其作用机理可能与减轻细胞膜损伤,降低线粒体膜通透性有关。Objective To observe the protective effects of β-asarone, an effective component of Rhizoma Acori Tatarinowii, on cultured neonatal rat myocardial cells with myocardial ischemia-reperfusion injury (MI-RI). Methods The primary neonatal rat myocardial cells were cultured, and the myocardial cells with MI-RI were induced by sodium dithionite. The survival rate of myocardial cells was detected by methyl thiazolyl tetrazolium (MTT) assay, contents of lactate dehydrogenase (LDH) and creatine kinase (CK) in the culture medium were detected with ultraviolet spectrophotometry, and the mean fluorescence intensity of mitochondrial membrane potential (MMP) was examined with flow cytometry (FCM). Results β-asarone increased the survival rate of MI-RI myocardial cells, decreased the contents of LDH and CK in the culture medium, and elevated MMP (P 〈 0.05). Conclusion β-asarone has protective effect on MI-RI myocardial cells, and its mechanism may be related with the reduction of cell membrane injury and the decrease of permeability of mitochondrial membrane.
