补阳还五汤对脊髓损伤大鼠脊髓组织血小板活化因子受体mRNA表达的影响

摘　　要：【目的】观察补阳还五汤对脊髓损伤(SCI)模型大鼠脊髓组织血小板活化因子受体(PAF-R)mRNA表达的影响。【方法】选用SPF级Wistar大鼠,随机分为4组:正常组、模型组、补阳还五汤组(剂量为40g·kg-·1d-1)、金纳多组(剂量为45.5mg·kg-.1d-1);采用Allens法复制中度SCI模型,造模成功后观察1周;连续给药2周后,取各组脊髓组织,采用实时逆转录聚合酶链反应(RT-PCR)法检测各组脊髓组织PAF-R mRNA表达水平。【结果】模型组大鼠脊髓组织PAF-R mRNA表达显著降低(P<0.01);补阳还五汤组及金纳多组均可抑制脊髓组织PAF-R mRNA表达,与模型组比较差异有显著性意义(P<0.01),而两给药组之间比较无显著性差异(P>0.05)。【结论】补阳还五汤对损伤脊髓的保护作用可能与其能减少PAF-R数目,抑制PAF-R活性,从而阻断PAF发挥损伤效应有关。Objective To observe the influence of Buyang Huanwu Decoction （BHD） on mRNA expression of platelet activating factor receptor （PAF-R） in spinal cord of rats with spinal cord injury （SCI）. Methods Specific-pathogen free Wistar rats were randomized into 4 groups： normal group, model group, BHD （40 g·kg^-1·d^- 1 ） group, and Ginaton （water solution of extract of Ginkgo Biloba leaves, 45.5 mg·kg^- 1·d^- 1 ） group. Rat models of moderate SCI were induced by Allen＇s method. The rats received corresponding medicine for 2 weeks after the modeling for one week. Then the spinal cord tissue was obtained, and PAF-R mRNA expression in spinal cord of different groups was observed by real-time reverse-transcription polymerase chain reaction （RT-PCR）. Results PAF-R mRNA expression in spinal cord of the model group was obviously decreased （P 〈 0. 01 ） , and BHD and Ginaton inhibited the spinal PAF-R mRNA expression （ P 〈 0. 01 compared with that in the model group）, but the difference between BHD group and Ginaton group was insignificant （P 〉 0. 05 ）. Conclusion The protective mechanism of BHD for spinal cord injury is probably related with the blockage of damage effect of PAF through reducing PAF-R number and inhibiting PAF-R activity.
