摘 要:水分胁迫(-1.2MPaPEG-6000)处理萌动的小麦种子,24h后诱导小麦幼芽产生41.5kD蛋白,其含量随着胁迫时间延长明显增加,48h时含量最高,到72h后不再变化。复水后,该蛋白消失;再胁迫48h时则又出现,其含量与处理24h时相当。41.5kD诱导蛋白主要位于细胞器膜上,细胞质中几乎不存在。41.5kD蛋白主要溶于10%NaCl提取液中,其等电点为pl5.65。该蛋白的氨基酸组成中,脯氨酸含量最高,其次为丙氨酸、天冬氨酸、谷氨酸、甘氨酸,没有发现半胱氨酸和组氨酸。Changs in protein profilesinduced by waer sbess (- 1. 2 MPaPEG) were investigated in shoots of ger-ndnating seeds of wheat. A protein(poforPtide) with an apmpnt MW of41 .5 kD was induced tmder waer sbess(Fig. 1 ). Its content increased with thehme of treatment. The level of 41. 5 kDprotein was enhanced significantly in the48 h of the beatmnt. Afer rewtering,the 41. 5 kD protein disappeared-Howwer, the 41. 5 kD protein appearedagain after rebeatmnt, and the level 0f4l. 5 kD protein was sindlar to tha aftertreatInnt of 24 h. The restilts of scans ofSDS-PAGE of proteins from the shoots ofwheat seediings showed that someoplypeptides disappeared, the content ofothers(e. g 36- 8 kD) increased andsome new polypeptides in the mp of l4to 25 kD were induced tmder watr sbess(Fig- 2). Delindnare results of subcel-lular forhonation indicated that the 41. 5kD protein was located dririly on or-ganelle membranes, almost none in cy-toso(Fig. 3 ). It was present Inainly inexusct with 10% NaCl(Fig.4). Analsisof itS theno acid composition sh0wd thatproline was the most abundant one, withAsp, Gly, Glu and kla in lesser amouns;no Cys and fus were found(Table 1 ).The isoelectric Point of 4l. 5 kD pmteinwas pI 5. 65.
