来氟米特联合环孢素A对大鼠心脏移植慢性排斥反应的抑制作用研究

摘　　要：目的探讨来氟米特对大鼠心脏移植慢性排斥反应抑制作用的机制,并评价来氟米特联合环孢素A对慢性排斥反应的作用效果。方法行大鼠腹部异位心脏移植后给予环孢素A 2.5 mg/(kg.d)灌胃,连续7天,以抑制急性排斥反应。第12天选跳动良好者随机入4组:对照组无治疗,来氟米特5 mg/(kg.d)组,环孢素A 5 mg/(kg.d)组;来氟米特5 mg/(kg.d)和环孢素A联合组。每组分别在42天、72天作组织病理学检查,RT-PCR检测血管内皮生长因子表达。结果与对照组相比,来氟米特组、环孢素A组及联合组对移植物炎性反应有明显抑制。联合组减少了VEGF表达,延缓了移植物新生血管病变进展。结论来氟米特与环孢素A联合用药组效果更佳,且能明显地抑制大鼠心脏移植血管病变的发展。Objective To discuss the mechanism of the leflunomide＇ s, inhibotany effect on chronic rejection in rat heart transplantation and evaluate the effect of the inhibition of leflunomide and cyclosporine A. Methods Rats underwent heterotopic heart transplantation in the abdomen. All rats were treated with cyclosporine A 2.5 mg/（ kg·d） for 7 days after operation to prevent acute rejection. On the 12 th day,40 recipients were divided into 4 groups at random： control group, leflunomide 5 mg/（kg·d）group, cyclosporine A 5 mg （kg·d）group and combination group. The cardiac grafts were harvested on the 42,72 th day after transplantation with HE stained and VEGF measured. Results The cardiac allograft vasculopathy process was delayed and the expression of VEGF was significantly lower in combination group. Conclusion The combination of leflunomide and cyclosporine A can inhibit the chronic rejection and reserve arterial intimal thickening and delay the process of cardiac allograft vasculopathy.
