角倍蚜与其唯一夏寄主植物盐肤木种群遗传多样性比较  被引量:4

摘  要:基于ISSR分子标记技术对贵州省8个种群共106个盐肤木样本进行遗传多样性和遗传分化分析,并与对应的角倍蚜种群ISSR研究结果进行比较.结果显示8个盐肤木种群11条引物随机扩增检测到171个多态位点,多态位点比率为100%,Shannon多样性指数和Nei基因多样性指数分析反映了与多态位点比率相一致的结果.该结果与角倍蚜多样性指数进行比较,发现角倍蚜和盐肤木种群的遗传多样性具有同样的变异趋势,且盐肤木种群的遗传多样性高于角倍蚜种群遗传多样性,相关性检验(P<0.01)表明二者之间具有明显的相关性;由Nei指数估计角倍蚜和盐肤木种群的遗传分化系数分别为0.23和0.19,二者具有相似的遗传分化水平,这种相似性表明二者具有明显的协同进化关系.UPGMA聚类结果表明,不同地域角倍蚜与盐肤木种群遗传距离与地理距离不相关,从二者聚类关系来看,也没有对应关系,这可能主要与二者属于明显不同的生物类群,以及本身固有的不同的生存方式和繁殖系统有关.The inter-simple sequence repeat (ISSR) maker was used to examine the genetic diversity and ge- netic differentiation of 8 Rhus chinensis populations from Guizhou Province, and meanwhile compared the genetic diversity with the Chinese gallnut aphid Schlechtendalia chinensis. R. chinensis is the unique prima- ry host of Chinese gallnut aphid. 11 ISSR primers were evaluated for their ability to produce polymorphic bands,and these primers yielded a total of 171 scorable bands, among which 100% were polymorphic. Shannon diversity index and Nei^s gene diversity has the similar results with the proportion of polymorphic loci. Compared the genetic index with that of S. chinensis and tested the correlation (P〈0.01) ,the results indicate that the diversity of R. chinensis has the same variation tendency with that of S. chinensis, and the genetic diversity of R. chinensis was higher than that of S. chinensis in case to the same population. Genetic differentiation coefficient was 22.94% and 19. 26% between R. chinensis populations and S. chinensis, showing the similar differentiation. This should be mainly lie in their coevolution relationship. Cluster anal- ysis showed that there was no direct correlation between genetic distance and geographic distance between them,and didn't form corresponding relation between populations, which maybe mainly result from the dif- ferent species with different reproduction system.
