摘  要:目的研究DNA跨损伤修复基因Revl、Polξ(Rev3和Rev7)、Polη、Polι、和Polk在脑胶质瘤中的表达,探讨这些基因的表达水平与脑胶质瘤发生、发展的关系。方法利用SYBRGreen实时定量PCR技术检测DNA跨损伤修复基因在85例原发脑胶质瘤病人和14名正常脑组织中的mRNA表达情况。结果与正常脑组织相比,Revl基因在Ⅳ级胶质瘤中表达上调(△Ct为5.55±1.12,P〈0.05);Rev3基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(△Ct分别为5.46±1.35、5.13±1.53和6.14±1.10,均P〈0.05);Rev7基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(△Ct分别为5.88±0.82、5.41±1.43和5.56±0.96,均P〈0.05);Polη基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(△Ct分别为5.97±1.15、5.83±1.03和6.34±1.0l,均P〈0.05);Polι基因在Ⅱ~Ⅳ级胶质瘤中表达上调(ACt分别为6.67±1.17、6.33±1.39和7.03±1.06,均P〈0.05);PolK基因在Ⅳ级胶质瘤中表达上调(△Ct为5.25±1.58,P〈0.05)。Spearman秩相关分析表明,跨损伤修复基因Revl、Rev3、Polm、PolL和PolK的表达量与肿瘤病理学分级呈正相关(均P〈0.01)。结论DNA跨损伤修复基因可能与胶质瘤的发生、进展相关。Objective To detect the expressions of Revl, Polξ, Polη, Pol ι and Pol K mRNA in human primary glioma tissues, and to analyze the relationship between the expression and the pathological grade. Methods We applied SYBR Green real-time PCR to investigate the mRNA expressions of the 6 genes in 85 primary glioma tissues and 14 normal brain tissues. Results The expressions of 4 genes including Rev3, RevT, Pol η and Pol ι were significantly ( P 〈 0. 05 ) higher in glioma tissues in grade Ⅱ , Ⅲ and Ⅳ as compared to normal brain tissues. The expressions of Revl and Pol K were significantly (P 〈 0. 05 ) higher in grade IV glioma compared to the normal brain tissue. The positive correlation existed ( P 〈 0. 01 ) between the expression of all genes mRNA and the pathologic grade of the human glioma, except for Rev7. Conclusion The up-regulation of DNA translesion synthesis genes, including Revl, Rev3, Rev7, Pol η, Pol ι and Pol K, may be associated with pathogenesis of glioma.
