摘  要:目的:优化栀子苷(Geniposide)乳酸-羟乙酸共聚物(PLGA)微球的制备工艺。方法:采用改进的复乳化-溶剂挥发法制备栀子苷PLGA微球,HPLC法测定微球的载药量及包封率,扫描电镜(SEM)观察微球的表面形貌,差示扫描量热(DSC)分析微球的包裹情况。结果:在优化条件下制备的栀子苷微球大小均匀,表面光滑圆整,载药量为6.15%,包封率为58.84%,DSC分析结果表明栀子苷能被PLGA材料有效包裹。结论:该制备工艺合理,为制备栀子苷PLGA微球提供了理论依据。AIM : To optimize the preparation process for the geniposide loaded poly lactic-co-glycolicacid ( PL- GA) microspheres. METHODS: A series of geniposide loaded PLGA microspheres have been prepared by W/o1/ o2 emulsion solvent evaporation method. The process parameters involved were investigated by HPLC with respect to the encapsulation and drug loading. The microspheres resulted with pellet appearance and the entrapment of micro- spheres were analysed by DSC. RESULTS: The obtained geniposide loaded PLGA microspheres had an average diameter showing smooth and round surface. The drug loading of microsphere was 6.15% and entrapment efficiency was 58.84%. Geniposide was successfully incorporated into PLGA microsphere through DSC analyses. CONCLUSION: The research provides an optimized method for the preparation of geniposide loaded PLGA microspheres.
