摘  要:目的研究白藜芦醇对糖尿病小鼠血脑屏障上P-糖蛋白(P-gp)表达和功能的影响。方法小鼠腹腔注射四氧嘧啶(ALX,200 mg.kg-1)诱导1型糖尿病。挑选空服血糖>11.1 mmol.L-1的小鼠分为糖尿病模型组、白藜芦醇低剂量组(7.5mg.kg-1)和白藜芦醇高剂量组(15 mg.kg-1)。另设正常对照组。白藜芦醇灌胃给药两周,用Western Blot技术测定血脑屏障上P-gp的蛋白水平;用罗丹明(Rh123)累积实验测定其P-gp的转运功能,并测定小鼠空腹血糖值(FSG)、糖化血红蛋白值(GHb)及脑内的活性氧(ROS)含量和总抗氧化能力(T-AOC)。结果白藜芦醇低剂量组和高剂量组小鼠血脑屏障上P-gp的表达水平显著升高,脑组织摄取Rh123水平(Kp)显著下降,同时白藜芦醇低剂量组和高剂量组的FSG和GHb明显低于模型组;脑组织总抗氧化能力显著高于模型组;而脑内ROS含量显著低于模型组。结论白藜芦醇能显著提高血脑屏障上P-gp表达水平和转运功能,这一作用可能与白藜芦醇降低脑内氧化应激水平有关。Aim To study the effects of resveratrol (Res) on the expression and function of p-gp at the BBB in the diabetic mice. Methods Alloxan (ALX, 200 mg·kg^-1) was injected intraperitoneally in mice to induce the type 1 diabetes model (T1DM) , among which the mice with over 11.1 mol·L^-1 of fasting serum glucose (FSG) were then randomly divided into three groups, namely the untreated T1DM group, the T1DM group treated with low dose Res (7.5 mg·kg^-1 ), the T1DM group treated with high dose Res ( 15 mg·kg^-1 ) , and the control group was set too. Two weeks after administration of the mice with Res, the p-gp' s expression at the BBB was examined by Western Blot, its transportation function was assayed by Rhodamine ( Rh123, a substrate of p-gp) accumulation test using florescence speetrophotometry. At the same time, the fasting serum glucose ( FSG), glycated hemoglobin ( GHb), the changes of reactive oxygen species (ROS) and the total antioxide capacity (T-AOC) of the mice brain were measured by eolorimetric method. Results The levels of the p-gp' s expression at the BBB were significantly higher and the capacities of brain uptake of Rh123 (Kp) were significantly lower in the T1DM group treated with low dose Res and high dose Res than those in the untreated T1DM group. In addition, compared with the untreated T1DM group, the levels of FSG and GHb were significantly decreased, and the total antioxide capacity (T-AOC) of the mice brain were remarkably increased and ROS levels were significantly decreased in the T1 DM groups treated with low dose Res and high dose Res. Conclusion Res could increase the expression and function of p - gp at the BBB in the diabetic mice, which may be involved in the reduction of the oxidative stress level in the brain of diabetic mice.
