成年大鼠脑神经干细胞表达PCNA和Ki-67抗原的比较研究

摘  要:目的比较成年大鼠侧脑室管膜下区(SVZ)和齿状回颗粒下区(SGZ)神经干细胞表达增殖细胞核抗原(PCNA)和Ki-67抗原的动态变化。方法采用免疫组织化学方法,分别对成年雄性SD大鼠SVZ和SGZ的PCNA阳性细胞及Ki-67阳性细胞进行定性和定量研究。结果PCNA或Ki-67免疫反应产物均位于细胞核,两种细胞的形态及分布也相似,但在SGZ或SVZ内的PCNA阳性细胞数均明显低于Ki-67阳性细胞数(P<0.05)。结论PCNA和K-67联合应用可以更全面和准确地评估成年在体神经干细胞的增殖状态。Objective To investigate the dynamic changes of proliferating cell nuclear antigen (PCNA) and Ki-67 antigen expression in the neural stem cells of subventricular zone (SVZ) of lateral ventricle and subgranular zone of dentate gyrus in adult rat brain. Methods Qualitative and quantitative analysis were made on the PCNA immunoreactive (PCNA-IR) ceils and Ki-67-IR cells both in SVZ and SGZ by means of immunohistochemistry. Results Both morphology and distribution patterns of PCNA-IR cells and Ki-67-IR cells were similar,but the number of positive Ki-67-IR cells in either SVZ or SGZ was significantly higher than that of PCNA-IR cells in either brain area (P 〈 0.05 ). Conclusion Combined application of both Ki-67 and PCNA is more valuable than single application of either antigen in detecting adult neurogenesis kinetics.
