摘  要:目的观察TNF-α(tumor necrosis factor-α)作用下人滑膜细胞RANKL(receptor activatornu clear factor kappa B ligand)及OPG(osteoprotegerin)mRNA表达的改变。方法体外原代培养获得人滑膜细胞,借助半定量RT-PCR(reverse transcription-polymerase chain reaction)检测细胞分别在不同浓度的TNF-α及其不同作用时段RANKL、OPGmRNA水平表达的变化。结果TNF-α能刺激人滑膜细胞RANKL和OPGmRNA的表达。同时,TNF-α浓度在10ng/mL时RANKL/OPG比值较对照组升高(P<0.01),并且TNF-α作用12h和24h时,RANKL/OPG比值均明显高于对照组(P<0.01)。结论TNF-α能够通过调节人滑膜细胞的RANKL/OPG的比值,从而可能参与影响病变关节周围骨破坏。[Objective] To investigate the effects of TNF-α on RANKL and OPG mRNA expressions in human synovial cells. [Method] The human synovial ceils were treated with TNF-α of different concentrations and periods. The mRNA expressions of RANKL and OPG were analyzed by RT-PCR. [Results] TNF-α could up-regulate the mRNA expression of both RANKL and OPG, while the RANKL/OPG ratio were upgraded at 10 ng/mL TNF-α con- centration. Moreover, the RANKL/OPG ratio was elevated significantly above the control group (P 〈0.01) at 12 and 24 hours. [Conclusion] TNF-α may affect osteoclasia around articulus which has pathological changes by up-regulating the RANKL/OPG expression ratio of synovial cells.
