摘  要:建立了微量沙眼衣原体组织培养法,并应用于不同人群的生殖道标本的沙眼衣原体培养。结果生殖道沙眼衣原体培养的阳性率:男性尿道炎患者147%(9/61);正常孕妇33%(7/210);急性和亚急性盆腔炎女性患者67%(7/104);性病门诊患者182%(49/269)。由于96孔培养板及倒置荧光显微镜的应用,微量培养法试剂用量少、过程简化;自制的抗沙眼衣原体单克隆抗体大大降低了培养后的染色成本,适用于大宗标本。Objectives: To isolate Chlamydia trachomatis by using a novel micro tissue culture. Design: Comparison of direct fluorescence antibody and indirct immunofluorescence stained Chlamydia trachomatis isolates. Methods: A micro tissue culture in which 96 well plate and inverted fluorescence microscope were used to isolate Chlamydia trachomatis from genital specimens of various populations. Results: The positive rate was found to be 14 7%(9/61) in men with urethritis, 3 3%(7/210)in pregnant women, 6.7%(7/104) in women with acute orsubacute pelvic inflammatory disease and 18.2%(49/269)in patients attending STD clinic. Procedure of the tissue culture was simplified by the use of 96 well plate and inverted fluorescence microscope and the cost was reduced due to the use of home made species specific monoclonal antibodies against the major outer membrane protein (MOMP) of Chlamydia trachomatis. Conclusion: The micro tissue culture by using 96 well plate and inverted fluorescence microscope and home made species specific monoclonal antibodies makes the culture procedure much simpler and more economic, especially for large scale samples.
