摘  要:目的:利用蛋白芯片技术和生物信息学方法从膝骨性关节炎患者血清中筛选标志蛋白质。方法:采用表面增强激光解吸电离飞行时间质谱技术(surfaced enhanced laser desorption/ionization time of flight mass spec-troscopy)和弱阳离子交换(CM10)蛋白芯片对30例肝肾亏虚型膝骨性关节炎患者、35例气滞血瘀型膝骨性关节炎患者血清和40例健康者血清的蛋白质谱图进行了检测,获得的蛋白质谱采用Ciphergen公司的Biomarker Wiz-ard和Biomarker Pattern软件分析。结果:肝肾亏虚型膝骨性关节炎患者与健康者血清蛋白质谱相比有18个显著差异蛋白质(P<0.01),其中13个蛋白质在患者血清中高表达,5个蛋白质在患者血清中低表达。气滞血瘀型膝骨性关节炎患者与健康者相比有20个显著差异蛋白质(P<0.01),其中6个蛋白质在患者血清中高表达,14个蛋白质低表达。肝肾亏虚型膝骨性关节炎患者与气滞血瘀型膝骨性关节炎患者相比有5个显著性差异蛋白(P<0.01)。结论:实验证明利用蛋白质组学和生物信息学方法可以从血清中筛选出膝骨性关节炎中医证型相关的标志蛋白质,而且蛋白质芯片技术对于发现和筛选血清中的膝骨性关节炎标志蛋白质是一种有效、快速的工具。Objective: New technologies for the study of knee osteoarthritis (OA) proteomic are urgendy needed. The aim of this study was to screen serum biomarkers in patients with knee OA by using surface - enhanced laser desorption and ionization time - of - flight mass spectrometry( SELDI - TOF - MS) technique. Methods :Proteomic spectra were gener- ated by mass spectrometry in 105 eases ,including 30 cases of deficiency of liver and kidney ,35 cases of stagnation of vital energy and blood stasis, and 40 cases of healthy control. The discriminatory profiling between knee OA and normal samples was analyzed by Biomarker Wizard software and Biomarker Pattern softwar. Results: eighteen differentially expressed proteins in serum were screened by analysis of protemie spectra of knee OA of deficiency of liver and kidney and healthy control ( P 〈 0.01 ). 13 proteins in knee OA of deficiency of liver and kidney were increased and 5 proteins in healthy controls were increased, twenty differentially expressed proteins in serum were screened by analysis of protemie spectra of knee OA of stagnation of vital energy and blood stasis and healthy control (P 〈 0.01 ). 6 proteins in knee OA of stagnation of vital energy and blood stasis were increased and 14 proteins in healthy controls were increased. 5 differentially expressed pro- teins in serum were screened by analysis of protemic spectra of knee OA of deficiency of liver and kidney and stagnation of vital energy and blood stasis( P 〈0.01 ). Concluion:SELDl -TOF- MS offers a unique platform for the proteomic de- tection of different symptom type of knee OA. It also offers a noninvasive method to further study the proteornic changes in the development and progression of knee OA.
