摘 要:目的探讨是否毛细血管扩张性共济失调(AT)细胞互补基因(ATDC)具有抑制AT细胞放射敏感性的作用和X线照射后AT细胞信号传导系统—PKCδ蛋白表达的改变。材料与方法LM217为SV40转化的正常人成纤维基质细胞。PB061为SV40转化的AT病人的成纤维基质细胞。实验分LM217和PB061±四环素(lμg/ml)3个组,分别照射不同剂量,求D0、α、SF2值。利用抗-PKCδ和抗PKCI1单抗进行免疫印迹杂交,PB061±四环素(1μg/ml)组分别照射0和5Gy。结果PB061+四环素(ATDC-)组的D0值为0.45Gy,α值为2.21Gy-1,SF2为0.01;而BP061(ATDC+)组的D0值为0.56Gy,α值为0.96Gy-1,SF2为0.10。其中α和SF2值统计学处理P<0.01,提示ATDC基因可以降低AT细胞的放射敏感性。Ca2+-非依赖蛋白激酶Cδ(PKCδ)在PB061细胞中表达水平较低,5Gy照射和四环素处理后均没有改变。PB061细胞中含有蛋白激酶C的抑制物(PKCI1),5Gy照射后和四环素处理后其蛋白表达没有改变。结论(1)ATDC基因具有抑?Purpose To investigate whether ATDC gene can lower the radiosensitivity of AT cells and influence the expression of PKCδ protein in AT Cells after ionizing radiation.Materials and Methods LM217 is a normal human fibroblast cell line transformed with SV40. PB061 is an AT Patient's fibroblast cell line transformed with SV40.The values of Do,α and SF2were measured by clone formation after various radiation doses on LM217,PB061± tetracycline(1μg/ml).WesternBlot analysis was performed using anti PKCδ and anti PKCI1 antibody to detect the expression of PKCδ and PKCI1 Protein. Results The values of Do,α and SF2 in PB061+ tetracycline(ATDC-) were 0.45Gy,2.21Gy-1 and 0.01 respectively; and those in PB061(ATDC+) were 0.56Gy,0.96Gy-1 and 0.10 respectively,the difference of α and SF2 value in PB061±tetracycline was significant.The expression of Ca2+independent PKCδ protein in PB061 was much less abundant,and did not show any change after 5Gy irradiation and tetracycline administration.The PKCI1 protein could be induced in PB061 cells and these gave no change in their expressive level after 5Gy irradiation and tetracycline administration.Conclusion (1)ATDC gene can lower the radiosensitivity of AT cells and the α value may be a major factor;(2) The PKCδ dose is not involved in the signal transduction pathway in PB061 cell after ionizing radiation.
