高效液相色谱法测定金银花中木犀草素含量

摘　　要：为了准确测定金银花中木犀草素的含量,通过摸索流动相、洗脱方式、保留时间和速度等条件,建立了高效液相色谱测定金银花中木犀草素含量的方法.采用SinoChrom ODS C18色谱柱(4.6 mm×150 mm,5μm),流动相为甲醇-0.3%H3PO4溶液,流速为0.8 mL/min,梯度洗脱.检测波长350 nm,柱温25℃,木犀草素的线性范围0.032 5～0.39μg/mL,回归方程为y=62.53x+10.88(r=0.999 0),平均回收率可达98.39%.测得金银花样品中木犀草素的含量为1.09%.In order to test the content of luteolin in honeysuckle, a quantitative analysis method of luteolin by HPLC was formed with MeOH-0.3%H3PO4 as the mobile phase on SinoChrom ODS C18 column（4.6 mm×150 mm, 5μm）. The flow-rate was 0.8 mL/min and the UV detection was set at 350 nm, with a column temperature of 25 ℃. The experimental results shows that the liner range of luteolin was 0.032 5-0.39 μg/mL with a correlation（r） of 0.999 0, a recovery rate of 98.39% and content rate of 1.09%.
