摘  要:目的了解妊娠中晚期和哺乳期接触磺胺二甲嘧啶对子代大鼠海马内神经生长因子(NGF)和脑源性神经营养因子(BDNF)的影响。方法清洁级SD母鼠在受孕第7日起至哺乳期结束分别每日喂饲磺胺二甲嘧啶50、100和200mg/kg,取其30日龄仔鼠的大脑进行免疫组化染色,观察海马内NGF和BDNF的表达。结果大脑的免疫组化分析发现,50、100、200 mg/kg组子代大鼠海马CA1和CA3区NGF表达的积分光密度值显著低于对照组(P<0.05),200 mg/kg剂量组BDNF表达的积分光密度值显著低于对照组(P<0.05)。结论磺胺二甲嘧啶喂饲染毒围生期母鼠,可能通过降低大鼠体内的甲状腺素浓度使脑组织NGF、BDNF表达下调,进而影响大脑的正常发育及功能。Objective To study the effect of perinatal exposure to sulphamethazine on the expression of neurotrophs (NGF and BDNF) in the hippoeampus of offspring in rats. Methods The dams (SD rats) were fed with sulphamethazine at the doses of 50, 100 and 200 mg/(kg·d) respectively from gestational day 7 to postpartum day 21. The brains of the pups aged 30 clays were collected and the expression of NGF and BDNF was determined by immunohistochemistry staining. Results The expression of NGF in CA1 and CA3 ofhippocampus significantly decreased in all pups of perinatal exposure to sulfamethazine (P〈0.05), and the expression of BDNF also significantly decreased in pups of perinatal exposure to 200 mg/(kg·d) sulphamethazine (P〈0.05). Conclusion Sulphamethazine can induce a down-regulated expression of NGF and BDNF by reducing TH level, which can affect the brain development.
