Effects of Lanthanum on Cell Multiplication and Intracellular Free Ca^(2+) Concentration in Primary Cultured Astrocytes of Rats

摘  要:目的观察氯化镧(LaCl3)对原代培养大鼠星形胶质细胞(astrocyte,AST)增殖能力和细胞内游离钙离子浓度的影响。方法AST经0.25、0.5、1.0和2.0 mmol/L LaCl3染毒24 h和48 h,四甲基偶氮唑盐(MTT)法检测细胞染LaCl3后增殖能力的变化;采用Fura-2/AM为荧光指示剂,F-4500型荧光双波长分光光度计测定细胞内游离钙离子浓度。结果细胞染毒24 h和48 h后,0.25、0.5、1.0和2.0 mmol/L LaCl3剂量组的细胞存活率分别为86.24%和82.99%,82.26%和79.14%,52.32%和49.97%及38.04%和35.15%,与对照组相比差异均有统计学意义(P<0.01);染毒24 h在倒置相差显微镜下观察,与对照组比较,1.0 mmol/L组细胞数量明显减少,胞体呈长梭形,胞突增粗,呈树枝状;2.0 mmol/L组上述改变加重,并可见大量脱壁细胞;在染毒24 h后,0.25、0.5、1.0 mmol/L LaCl3剂量组细胞内Ca2+浓度分别为(134.41±0.83)、(163.31±3.97)、(224.84±5.51)nmol/L,显著高于对照组[(129.45±1.09)nmol/L](P<0.05)。结论LaCl3体外染毒可降低AST的增殖能力,其机制可能与其所致的细胞内Ca2+浓度超载有关。Objective To study the effects of lanthanum on cell multiplication and intracellular Ca^2+([Ca^2+) concentration in primary cultured astrocytes of rats. Methods The primary cultured astrocytes of rats were exposed to 0.25, 0.5 , 1.0 and 2.0 mmol/L LaCI3 for 24 h and 48 h.The cell multiplication was measured by MTT; [Ca^2+]i concentration was determined by the fluorescent Ca^2+ indicator Fura-2/AM. Results After astrocytes were incubated with 0.25,0.5,1.0 and 2.0 mmol/L LaCl3 for 24 h and 48 h, the survival rate of astrocytes was 86.24% and 82.99%, 82.26% arid 79.14%, 52.32% and 49.97%, 38.04% and 35.15% respectively, which was significantly decreased compared with the control group ( P〈0.01). Compared with the control group, the quantity of astrocytes was decreased significantly after incubated with 1.0 mmol/L LaCl3, and meanwhile the cell bodies presented on fusiform shape and the neurodendrites were thickening under the inverted phase contrast microscope. Compared with the control group, [Ca^2+]i respectively increased from (129.45±1.09)nmol/L to (134.41±0.83)nmol/L (163.31±3.97)nmol/L, (224.84±5.51)nmol/L after astrocytes were incubated with 0.25,0.5 and 1.0 mmol/L LaCl3 for 24 h (P〈0.05). Conclusion Lanthanum exposure may decrease the astrocytes multiplication in vitro, which is probably related to the overload of [Ca^2+]i.
