摘  要:目的探讨在铜摄入量低于正常摄入量的情况下血中铜、锌含量、铜/锌比值及血脂的相关关系,为铜的多途径暴露提供统一的剂量依据。方法42只SD大鼠随机分成7组,其中1组作为正常对照组喂饲正常饲料、饮用正常自来水;另外6组喂饲缺铜饲料、饮用去离子水,同时分别给予不同剂量葡萄糖酸铜,依次为缺铜模型对照组(0ADI,0 mg/ml)、1/625ADI(0.000128 mg/ml)、1/125ADI(0.000 64 mg/ml)、1/25ADI(0.003 2 mg/ml)、1/5ADI(0.016 mg/ml)及ADI(0.8 mg/ml)组,30 d后测定其血中铜蓝蛋白(CP)活力作为判断机体铜缺乏标准,测定其血中铜、锌含量及血中总胆固醇(TC)、甘油三脂(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL),进行相关性研究。结果铜摄入量不足时,各组大鼠血中铜蓝蛋白活力低于正常水平(P<0.05),说明机体处于铜缺乏状态。此时,除1/25ADI组外,其他各组血铜值变化不明显而各组血中铜/锌比值(Cu/Zn值)均高于正常水平(P<0.05),且缺铜模型组、1/625ADI组TC、HDL、LDL水平均明显低于正常对照组(P<0.05)。在1/25 ADI组血铜及Cu/Zn值均出现异常升高(P<0.05),而TC、HDL、LDL值异常降低(P<0.05)。缺铜模型组大鼠血中TG水平高于正常水平(P<0.05),当有少量铜摄入后,1/625ADI、1/125ADI及1/25ADI各组TG水平明显低于正常对照组(P<0.05),且随着铜摄入量的升高而升高。结论铜摄入量低于日常摄入量时,机体处于铜缺乏状态,此时血中Cu/Zn值反应比血铜更为敏感。缺铜会导致血中TC、TG、HDL、LDL低于正常水平,而在1/25ADI时TC、HDL、LDL异常降低,0 mg/ml铜摄入量组TG异常升高。Objective To study the relation between copper, copper-zinc ratio in blood and blood-fat only in the copper intake lower than the normal acceptable daily intake, and to provide a unified dosage evidence for copper multi-ways exposure. Methods Forty-two SD rats were divided into 7 groups randomly. The rats in one group among them as the normal control group were fed on the normal fodder and water, the other 6 groups were fed on special fodder without copper and deionized water and i.g. copper gluconate at doses of 0ADI (the acceptable daily intake of rat), 1/625ADI (0.000 128 mg/ml), 1/125ADI (0.000 64 mg/ml), 1/25ADI (0.003 2 mg/ml), 1/SADI (0.016 mg/ml) and ADI (0.8 mg/ml), for 30 days. The activity of ceruloplasmin (CP), the content of copper, zinc in the blood and the level of total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL) and low densitylipoprotein (LDL) were determined. Results In the case of copper intake lower than the normal acceptable daily intake, the activity of CP in the blood of all treated rats were lower than the normal level (P〈0.05), suggested the body were in the case of copper deficiency. The change of the content of copper were no-obvious while the copper-zinc ratio (Cu/Zn) were higher than the normal level (P〈0.05). The level of TC, HDL and LDL of no copper model group and 1/625ADI were obviously lower than the normal level (P〈0.05). In the 1/25ADI groups, the copper content and Cu/Zn increased abnormally (P〈0.05) leading to the abnormal decrease of the TC, HDL and LDL level (P〈0.05). As for the level of TG, the no copper model group was higher than the normal level (P〈0.05), while turned into lower than the normal level (P〈0.05) when the copper intake were in 1/625ADI, 1/125ADI and 1/25ADI.
