摘 要:目的观察中药复方益肾蠲湿合剂对肾小球系膜细胞(GMC)分泌细胞外基质的抑制作用。方法将含低、中、高剂量益肾蠲湿合剂的药物血清和10%血清-DMEM培养液共同培养GMC 24,48和72 h,分别收集细胞培养上清,用Elisa法检测培养上清中纤维连结蛋白(FN)、层黏连蛋白(LN)及Ⅳ型胶原(ColⅣ)含量。结果益肾蠲湿合剂的药物血清可抑制体外培养的GMC分泌FN、LN和ColⅣ,其抑制程度与药物浓度有一定的量效关系,即随药物浓度的增加对GMC分泌FN、LN和ColⅣ的抑制率也相应增加。结论中药复方益肾蠲湿合剂对体外培养的GMC分泌FN、LN和ColⅣ有明显的抑制作用,提示抑制GMC分泌细胞外基质是本复方治疗慢性肾炎的重要机制之一。Objective It is to observe the inhibiting effect of Yishenjuanshi decoction on the secretion of extracelluar matrix in cultured glomerular mesangial cells (GMCs). Methods GMC were cultured in cultured solutions contained low, medi um and high dose of Yishenjuanshi decoction respectively with 10 % serum-DMEM for 24 h, 48 h and 72 h. After that, cell supernatant was collected to determine the concentrations of fibronectin (FN), laminin (LN) and collagen Ⅳ (Col Ⅳ ) in it with Elisa method. Results Drug serum contained Yishenjuanshi could significantly inhibit GMC cultured in vitro to secrete FN, LN and ColⅣ. There was a dose-effect relationship between the inhibiting degree and drug concentration. The inhibi- tion ratios increased accordingly when the dose of the decoction increased. Conclusion Yishenjuanshi has an obvious inhibiting function on secretion of FN, LN and ColIV of GMC cultured in vitro. It indicates that inhibiting GMC to secret extracelluar matrix is one of the important mechanisms of this decoction treating for chronic nephritis.
